Network Perturbation Analysis Identifies Three Molecular Subtypes With Distinct Clinical Outcomes and Therapeutic Vulnerabilities in Endometrial Cancer

open access: yesiMetaMed, EarlyView.
We performed network perturbation analysis on transcriptomic data from endometrial cancer patients across multiple cohorts and identified three molecular subtypes with distinct clinical outcomes. The most aggressive subtype, C1, exhibits comprehensive molecular and immunological alterations, including dysregulated hormone signaling, immune exclusion ...

Dynamics and Regulation of Maternal–Fetal Interface Macrophages: From Immune Tolerance to Pregnancy Outcomes

open access: yesiMetaMed, EarlyView.
The maternal–fetal interface demands tolerance alongside antimicrobial defense. Plastic macrophages orchestrate inflammation resolution, tissue remodeling, and angiogenesis in normal gestation; complications (miscarriage, preterm birth) feature polarization skewing and dysfunction.

Homoharringtonine suppresses acute myeloid leukemia progression by orchestrating EWSR1 phase separation in an m6A‐YTHDF2‐dependent mechanism

open access: yesiMeta, EarlyView.
Homoharringtonine (HHT), a clinically approved anti‐leukemic drug, directly binds the RNA‐binding protein EWSR1. HHT induces an allosteric conformational switch in the RNA recognition motif of EWSR1, driving self‐association and liquid–liquid phase separation (LLPS). The resulting condensates act as regulatory hubs that sequester the m6A reader YTHDF2,
