Endothelin and endothelin receptor antagonism in portopulmonary hypertension

European Journal of Clinical Investigation, 2006
AbstractPortopulmonary hypertension (PPHT) is a rare but devastating complication in patients with portal hypertension, characterized by pulmonary arterial obliterative disease with a concomitant rise in pulmonary vascular resistance. A broad body of evidence has accumulated, indicating that endothelin (ET) peptides and their cognate receptors are ...

Endothelin Receptor Antagonists

2013
Three pathways have been identified in the pathogenesis of pulmonary arterial hypertension (PAH): the endothelin (ET), nitric oxide (NO) and prostacyclin pathways. These pathways represent the targets of approved PAH therapies and their discovery has facilitated significant progress in the understanding and treatment of PAH.

Histochemical Studies on Endothelin and the Endothelin-A Receptor in the Hypothalamus

Journal of Cardiovascular Pharmacology, 1998
By use of a rabbit polyclonal antibody specific to the endothelin-A (ETA) receptor, we analyzed the distribution of neurons containing ETA receptors in rat brain. Almost all A1-A7 noradrenergic neurons, A8-A16 and retinal amacrine (A17) dopaminergic neurons (except the A15 group), and C3 adrenergic neurons contained ETA receptors.

Internalization of Type-A Endothelin Receptor

Journal of Cardiovascular Pharmacology, 2000
Endothelins (ETs) are 21 amino acid peptides which bind to ET(A)- and ET(B)-receptors to evoke diverse physiological responses. This report studies the internalization of ET(A)-receptor in Chinese hamster ovary (CHO) cells which were stably transfected with ET(A)-receptor cDNA.

Endothelins and their receptors in embryo implantation

Journal of Cellular Biochemistry, 2019
AbstractAs a critical stage of pregnancy, the implantation of blastocysts into the endometrium is a progressive, excessively regulated local tissue remodeling step involving a complex sequence of genetic and cellular interplay executed within an optimal time frame. For better understanding the causes of infertility and, more importantly, for developing

Endothelin and Endothelin Receptor Antagonists in Heart Failure

Congestive Heart Failure, 2002
Endothelin (ET) is a recently discovered 21‐amino acid peptide that has potent physiologic and pathophysiologic effects that appear to be involved in the development of heart failure. These include effects on arterial smooth muscle cells that cause intense peripheral vasoconstriction and stimulation of cardiac myocytes and fibroblasts.

Enrasentan, an Antagonist of Endothelin Receptors

Cardiovascular Drug Reviews, 2003
ABSTRACTEndothelins are powerful vasoconstrictor agents produced by endothelial cells and identified by Yanagisawa et al. in 1988. Two types of receptors for endothelins have been identified: ETAreceptors are located on smooth muscle cells of the vascular wall and are responsible for endothelin‐induced vasoconstriction while ETBreceptors are located on

Endothelin Receptor Antagonists - An Overview

Current Medicinal Chemistry, 2002
In thirteen years since the appearance of Endothelin (ET) on the international scene, possibility of its involvement in a variety of diseases has attracted the attention of medicinal chemists in search of novel therapeutics for various cardiovascular diseases (CVDs).

Endothelin Receptors and Receptor Subtypes

1992
Endothelin, originally isolated from culture media of porcine aortic endothelial cells, is one of the most potent vasoactive peptide known to date and consists of 21 amino acids containing two intracellular disulfide bonds.21 In addition to vasoconstrictor activity, it has been found that endothelin has a variety of biological actions: it increases ...

Effect of castration on endothelin receptors

Clinical Science, 2002
Endothelin (ET) plays a pivotal role in the pathogenesis of cell growth disorders such as cancer. Atrasentan (ABT-627), a selective antagonist for the ET receptor A (ETA), has shown benefit in controlling disease progression in men with hormone refractory prostate cancer who have undergone aggressive hormone ablation therapy.
