Structural insights into an engineered feruloyl esterase with improved MHET degrading properties

open access: yesFEBS Letters, EarlyView.
A feruloyl esterase was engineered to mimic key features of MHETase, enhancing the degradation of PET oligomers. Structural and computational analysis reveal how a point mutation stabilizes the active site and reshapes the binding cleft, expading substrate scope.

Duality origami: Emergent ensemble symmetries in holography and Swampland

open access: yesPhysics Letters B
We discuss the interrelations between several ideas in quantum gravity – holography, the Swampland, and the concept of ensemble averaging. To do so, we study ensemble averages of Narain-type theories associated with general even quadratic forms and their

The perceptual average in ensemble representation: Neither perceptual nor an average. [PDF]

open access: yesAtten Percept Psychophys
Abstract This report targets the claim that gist representations of visual stimuli, called “ensemble averages”, are perceptual representations of statistics pertaining to stimuli. We report predictions of a mathematical model based on classical memory architectures which assumes ensemble averages are statistical approximations to ...

A Note on Functional Averages over Gaussian Ensembles [PDF]

open access: yesJournal of Probability and Statistics, 2013
We find a new formula for matrix averages over the Gaussian ensemble. LetHbe ann×nGaussian random matrix with complex, independent, and identically distributed entries of zero mean and unit variance. Given ann×npositive definite matrixAand a continuous functionf:ℝ+→ℝsuch that∫0∞‍e-αt|f(t)|2dt<∞for everyα>0, we find a new formula for the ...

Modulation of Homer1 EVH1 domain internal dynamics by putative autism‐associated mutations

open access: yesFEBS Letters, EarlyView.
The putative autism‐associated M65I and S97L variants of the EVH1 domain of the postsynaptic scaffold protein Homer1 do not exhibit substantial changes in their overall structure or partner binding. Both of them, but especially the M65I variant, show altered internal dynamics relative to the wild‐type domain on the μs‐ms timescale, indicated by the ...

Salmonella lipopolysaccharide‐containing supported lipid bilayers as platforms to study bacteriophage interactions

open access: yesFEBS Letters, EarlyView.
We present robust protocols for the preparation of supported lipid bilayers (SLBs) incorporating either Salmonella smooth LPS or outer membrane vesicles (OMVs). We use a combination of quartz crystal microbalance with dissipation (QCM‐D) and fluorescence microscopy to both characterize the SLBs of various compositions and to probe their interactions ...

Investigating transcription factor dynamics in health and disease using FRAP

open access: yesFEBS Letters, EarlyView.
FRAP analysis of GFP‐tagged transcription factors reveals how molecular mobility and target engagement change in response to drug treatment. By combining live‐cell imaging, quantitative model fitting, and statistical analysis, this approach uncovers transcription factor dynamics linked to disease mechanisms, providing a powerful framework for ...

Closed universes, factorization, and ensemble averaging

open access: yesJournal of High Energy Physics
We study closed universes in holographic theories of quantum gravity. We argue that within any fixed theory, factorization implies there is one unique closed universe state.

Single‐cell DNA methylation profiling: Technologies, computation, and applications in precision oncology

open access: yesMolecular Oncology, EarlyView.
Single‐cell DNA methylation (scDNAme) profiling maps epimutational clonal evolution, revealing mechanisms of malignancy and therapeutic resistance across diverse cancer types. By providing a high‐resolution landscape of intratumoral heterogeneity, these technologies empower precise patient stratification, guide the development of enhanced ...

A light‐triggered Time‐Resolved X‐ray Solution Scattering (TR‐XSS) workflow with application to protein conformational dynamics

open access: yesFEBS Open Bio, EarlyView.
Time‐resolved X‐ray solution scattering captures how proteins change shape in real time under near‐native conditions. This article presents a practical workflow for light‐triggered TR‐XSS experiments, from data collection to structural refinement. Using a calcium‐transporting membrane protein as an example, the approach can be broadly applied to study ...
