Advanced Science, EarlyView.Gemcitabine resistance remains a major challenge in pancreatic ductal adenocarcinoma. This study identifies STYK1 as a key mediator of chemoresistance through autophagy‐dependent maintenance of cancer stemness. Gemcitabine‐activated AMPK phosphorylates STYK1 at Ser91 and Ser304, enhancing autophagy and stemness.
