Quantitative Visualization of Intracellular Nanometabolism Using Peak‐Shifted Dual‐State Emissive FRET Nanoprobes

This study reveals the long‐elusive intracellular dissolution mechanism of carrier‐free nanomedicines. By utilizing peak‐shifted dual‐state emissive FRET nanoprobes (PDFNPs) that undergo a distinct ratiometric fluorescence peak shift upon disassembly, we achieve real‐time, quantitative tracking of dissolution kinetics in live cells.

Discovery and Optimization of AMT‐676, a CDH17‐Targeting ADC for the Treatment of Advanced Gastrointestinal Cancers

AMT‐676 is a novel antibody‐drug conjugate targeting CDH17, an adhesion molecule uniquely exposed on gastrointestinal tumor surfaces. Engineered with an optimized exatecan payload, it demonstrates profound tumor regression and a robust bystander effect across diverse preclinical models.

Single‐Molecule Imaging of Endogenous RNA Using a Miniaturized and Circularly Permuted CRISPR‐Cas13 Degron‐Masking System CDegSR

CDegSR, a new tool for imaging RNA at single molecule level in living cells was developed by researchers. Background noise was eliminated by protein engineering and making it degrade when not bound to its target. This allowed individual RNA molecules to be clearly imaged and tracked.
