Engineering Osteoimmune Responses with Functionalized Orthopedic Implants for Post‐Operative Osteosarcoma Treatment

open access: yesAdvanced Science, EarlyView.
Osteosarcoma is the most common primary bone tumor with limited treatment options and a terrible prognosis. This review provides a comprehensive summary of the recent development of osteoimmunomodulatory implants for post‐operative osteosarcoma treatment, of which the potential utility in evoking durable anti‐osteosarcoma immunity and accelerating bone

Combined Photothermal and mTOR‐Targeted Therapy Overcomes Immune Evasion and Enhances Checkpoint Blockade Efficacy in Metastatic Triple‐Negative Breast Cancer

open access: yesAdvanced Science, EarlyView.
This study reveals that photothermal therapy, while inducing immunogenic cell death in triple‐negative breast cancer, paradoxically activates the oncogenic mTOR pathway to drive immune evasion. To counter this, a smart nanocomposite is engineered to co‐deliver localized hyperthermia and mTOR inhibition.

Inulin‐Based Oral Chemotherapy Modulates Gut Microbiota and Immune Microenvironment through Inhibition of Neutrophil Extracellular Trap Formation for Improving Cancer Therapy

open access: yesAdvanced Science, EarlyView.
Schematic representation of ZIF‐8@OXA@inulin for cancer immunotherapy. Synthesis of ZIF‐8@OXA@inulin. Augmentation of pyroptosis, regulation of gut microbiota, and inhibition of NETs formation by ZIF‐8@OXA@inulin‐mediated comprehensive strategy for antitumor immune response. ABSTRACT Despite the potential of chemoimmunotherapy against colorectal cancer

PRDM1+ Malignant Cells Mediate an Immunosuppressive Landscape and Resistance to Neoadjuvant Chemoradiotherapy and Immunotherapy in Esophageal Squamous Cell Carcinoma

open access: yesAdvanced Science, EarlyView.
Integrated scRNA‐seq, scTCR‐seq analysis, and functional assays identify PRDM1+ malignant epithelial cells with hyper lipid peroxidation characteristics that demonstrate reduced responsiveness to the nICRT treatment. Principal factor PRDM1 activates cysteine metabolism genes to modulate lipid peroxidation (an intrinsic cellular pathway related to ...

Investigating the determinants of fear of cancer recurrence in patients with early‐stage non‐small cell lung cancer: Insights from a restricted cubic spline model

open access: yesCancer Medicine
Objective This study aims to investigate the determinants and the dose–response relationship of fear of cancer recurrence among patients with early‐stage non‐small cell lung cancer (NSCLC), aiming to inform prevention and intervention strategies. Methods

Targeting Lactate and Lactylation in Cancer Metabolism and Immunotherapy

open access: yesAdvanced Science, EarlyView.
Lactate, once deemed a metabolic waste, emerges as a central regulator of cancer progression. This review elucidates how lactate and its epigenetic derivative, protein lactylation, orchestrate tumor metabolism, immune suppression, and therapeutic resistance.

pH/ROS‐Responsive Injectable Hydrogel Co‐Loaded with B7‐H3 Blocker and NETs Suppressor Boosts OSCC Synergistic Immunotherapy

open access: yesAdvanced Science, EarlyView.
A pH/ROS‐dual‐responsive injectable hydrogel, formed by dynamic boronic ester and Schiff base crosslinking, co‐delivers enoblituzumab (B7‐H3 blocker) and Cl‐amidine (NETs suppressor). Via localized intratumoral delivery, it synergistically reprograms the immunosuppressive tumor microenvironment, overcomes T cell infiltration barriers, restores ...

EGR Proteins Mediate Interferon‐Independent Anti‐HSV‐1 Responses Through Viral and Host Targets

open access: yesAdvanced Science, EarlyView.
Early antiviral responses are typically mediated by interferons. However, during HSV‐1 infection, host early growth response (Egr) genes, which are not interferon‐stimulated genes, are quickly induced by viral protein ICP0. EGR proteins, in turn, suppress viral lytic infection by activating viral latency‐associated (LAT) and host immune regulatory ...

Calcium Channel Blockers Inhibit Pancreatic Neuroendocrine Neoplasms Progression via Cav1.2‐Epigenetic Circuit

open access: yesAdvanced Science, EarlyView.
Our study reveals a novel mechanism of a positive regulatory circuit between Cav1.2 and H3K27ac for pancreatic neuroendocrine neoplasms (pNENs) progression. Cav1.2 is identified as a crucial target for promoting disease progression and correlates with malignant behaviors, which are remarkably inhibited by the administration of calcium channel blockers (
