Fibrous dysplasia (FD) is caused by somatic mutations in GNAS and activation of the cAMP-regulating protein, Gs alpha ( gsp ). gsp inhibits differentiation of stem cells to mature osteogenic cells, resulting in replacement of normal bone with a highly proliferative stroma composed of cells that express osteogenic markers. Apoptosis follows, leaving

Fibrous dysplasia of bone is a congenital non hereditary benign bone disease, where normal bone is replaced by a fibrous-like tissue with immature osteogenesis. Prevalence is difficult to estimate, due to frequent asymptomatic lesions. Bone lesions are mono- or polyostotic and may be associated with bone pain and fragility, leading to fractures.

At hand of 5 clinical and pathological observations, the Authors report on the main features of fibrous Bone Dysplasia. Roentgenological and histological appearances are illustred. In some polyostotic and roentgenologically atypical forms the diagnosis must be histologically confirmed.
