Better than a lens -- Increasing the signal-to-noise ratio through pupil splitting

open access: yes, 2022
Lenses are designed to fulfill Fermats principle such that all light interferes constructively in its focus, guaranteeing its maximum concentration. It can be shown that imaging via an unmodified full pupil yields the maximum transfer strength for all ...

An isoform of 14‐3‐3 protein regulates transbilayer lipid movement at the plasma membrane

open access: yesFEBS Letters, EarlyView.
Loss of 14‐3‐3ζ in CHO cells confers resistance to exogenous phosphatidylserine (PS) and impairs endocytosis‐independent inward flip‐flop of fluorescent PS at the plasma membrane. RNAi‐mediated knockdown reproduces this defect, while no additive effect is seen in ATP11C‐deficient cells.

Virtual-freezing fluorescence imaging flow cytometry

open access: yesNature Communications, 2020
High throughput imaging flow cytometry suffers from trade-offs between throughput, sensitivity and spatial resolution. Here the authors introduce a method to virtually freeze cells in the image acquisition window to enable 1000 times longer signal ...

$\Lambda$-Enhanced Imaging of Molecules in an Optical Trap

open access: yes, 2018
We report non-destructive imaging of optically trapped calcium monofluoride (CaF) molecules using in-situ $\Lambda$-enhanced gray molasses cooling. $200$ times more fluorescence is obtained compared to destructive on-resonance imaging, and the trapped ...

Fluorescence live cell imaging [PDF]

open access: yes, 2014
Fluorescence microscopy of live cells has become an integral part of modern cell biology. Fluorescent protein (FP) tags, live cell dyes, and other methods to fluorescently label proteins of interest provide a range of tools to investigate virtually any cellular process under the microscope.

The ubiquitin ligase RNF115 is required for the clearance of damaged lysosomes

open access: yesFEBS Letters, EarlyView.
Upon lysosomal rupture, an E3 ubiquitin ligase RNF115 translocates from the cytosol to the damaged lysosomal membrane. Moreover, RNF115 depletion impairs the clearance of damaged lysosomes, identifying it as a key regulator of lysosomal quality control.

Harmonized technical standard test methods for quality evaluation of medical fluorescence endoscopic imaging systems

open access: yesVisual Computing for Industry, Biomedicine, and Art
Fluorescence endoscopy technology utilizes a light source of a specific wavelength to excite the fluorescence signals of biological tissues. This capability is extremely valuable for the early detection and precise diagnosis of pathological changes ...

Improved fluorescent phytochromes for in situ imaging

open access: yesScientific Reports, 2022
Modern biology investigations on phytochromes as near-infrared fluorescent pigments pave the way for the development of new biosensors, as well as for optogenetics and in vivo imaging tools.

AAA+ protein unfoldases—the Moirai of the proteome

open access: yesFEBS Letters, EarlyView.
AAA+ unfoldases are essential molecular motors that power protein degradation and disaggregation. This review integrates recent cryo‐electron microscopy (cryo‐EM) structures and single‐molecule biophysical data to reconcile competing models of substrate translocation.

Degradation in Field-aged Crystalline Silicon Photovoltaic Modules and Diagnosis using Electroluminescence Imaging [PDF]

open access: yes, 2016
Degradation phenomena observed in field-aged crystalline silicon photovoltaic modules include EVA browning, delamination between the glass-encapsulant and the cell-encapsulant interfaces, degradation of the anti-reflective coating, corrosion of busbars ...
