Single-pixel phase and fluorescence microscope

open access: yesOptics Express, 2018
Multimodal microscopes either use multiple cameras or a single camera to multiplex different modes spatially. The former needs expertise demanding alignment and the latter suffers from limited spatial resolution. Here, we report an alignment-free full-resolution simultaneous fluorescence and phase imaging approach using single-pixel detectors.

CDK11 inhibition induces cytoplasmic p21WAF1 splice variant by p53 stabilisation and SF3B1 inactivation

open access: yesMolecular Oncology, EarlyView.
CDK11 inhibition stabilises the tumour suppressor p53 and triggers the production of an alternative p21WAF1 splice variant p21L, through the inactivation of the spliceosomal protein SF3B1. Unlike the canonical p21WAF1 protein, p21L is localised in the cytoplasm and has reduced cell cycle‐blocking activity.

Improving PARP inhibitor efficacy in bladder cancer without genetic BRCAness by combination with PLX51107

open access: yesMolecular Oncology, EarlyView.
Clinical trials on PARP inhibitors in urothelial carcinoma (UC) showed limited efficacy and a lack of predictive biomarkers. We propose SLFN5, SLFN11, and OAS1 as UC‐specific response predictors. We suggest Talazoparib as the better PARP inhibitor for UC than Olaparib.

Cell sorting in a Petri dish controlled by computer vision. [PDF]

open access: yes, 2013
Fluorescence-activated cell sorting (FACS) applying flow cytometry to separate cells on a molecular basis is a widespread method. We demonstrate that both fluorescent and unlabeled live cells in a Petri dish observed with a microscope can be ...

Cytoplasmic p21 promotes stemness of colon cancer cells via activation of the NFκB pathway

open access: yesMolecular Oncology, EarlyView.
Cytoplasmic p21 promotes colorectal cancer stem cell (CSC) features by destabilizing the NFκB–IκB complex, activating NFκB signaling, and upregulating BCL‐xL and COX2. In contrast to nuclear p21, cytoplasmic p21 enhances spheroid formation and stemness transcription factor CD133.

Self-confocal NIR-II fluorescence microscopy for multifunctional in vivo imaging

open access: yesJournal of Innovative Optical Health Sciences
Fluorescence imaging in the second near-infrared window (NIR-II, 900–1880[Formula: see text]nm) with less scattering background in biological tissues has been combined with the confocal microscopic system for achieving deep in vivo imaging with high ...

Enhancement of Fluorescent Labeling via a Composited Thin Film

open access: yesInternational Journal of Polymer Science, 2014
Fluorescent labeling is the prevailing imaging technique in cell biological research. When statistical investigations on a large number of cells are involved, experimental study is required for both low magnification to get a reliable statistical ...

Build Your Own Microscope: Step-By-Step Guide for Building a Prism-Based TIRF Microscope

open access: yesMethods and Protocols, 2018
Prism-based total internal reflection fluorescence (pTIRF) microscopy is one of the most widely used techniques for the single molecule analysis of a vast range of samples including biomolecules, nanostructures, and cells, to name a few.

Fibrillisation of hydrophobically modified amyloid peptide fragments in an organic solvent [PDF]

open access: yes, 2007
The self-assembly of a hydrophobically modified fragment of the amyloid beta(A beta) peptide has been studied in methanol. The peptide FFKLVFF is based on A beta(16-20) extended at the N terminus by two phenylalanine residues.

Class IIa HDACs forced degradation allows resensitization of oxaliplatin‐resistant FBXW7‐mutated colorectal cancer

open access: yesMolecular Oncology, EarlyView.
HDAC4 is degraded by the E3 ligase FBXW7. In colorectal cancer, FBXW7 mutations prevent HDAC4 degradation, leading to oxaliplatin resistance. Forced degradation of HDAC4 using a PROTAC compound restores drug sensitivity by resetting the super‐enhancer landscape, reprogramming the epigenetic state of FBXW7‐mutated cells to resemble oxaliplatin ...
