Calibrating fluorometers for clinical diagnostics

Everyone would like to calibrate their fluorometers with a stable transferable fluorescent object. Existing stable fluorophores generally do not match the geometrical, spectral or anisotropic properties of samples used in in-vitro diagnostics. This paper will discuss the ideal features of fluorescence calibrators and why no practical calibrators exist.

Single-turnover saturating flashes of light are used to induce synchronous operation of Photosystem II reaction centers. The flash must be so short that the frequency of double-turnovers is negligible and must be powerful enough to turnover all reaction centers.

Fluorometer Calibration Curves and Nomographs

The relationship of rhodamine B dye fluorescence to water temperature is confirmed, and a procedure for calibrating a fluorometer for time-of-travel dye studies is described. Calibration nomographs for the four aperture settings designed into fluorometers are developed from experimentally derived temperature-fluorescence curves.
