Microchimica ActaAn aptamer-antibody sandwich electrochemical immunosensor was studied. Fe3O4/MWCNTs-COOH/Nafion was modified and fixed on a glassy carbon electrode to amplify electrical signals. The antibody was coupled with AuNPs to form conjugates. It captures the target free thyroxine (FT4) and forms antibody-AuNPs/FT4 macromolecular complexes.

The assay comprises a two-vial-procedure to measure total thyroxine and free thyroxine fraction by use of monoclonal antibodies. Free thyroxine concentrations are calculated from fT4-fraction and total thyroxine concentration readings.

In 211 consecutive patients T4/TBG ratio correlated well with direct FT4 determination over a wide range of TBG concentration. However, there was a tendency to decreasing T4/TBG ratios with an increase of TBG concentration, which in consequence did lead to false results in three patients with very high TBG concentrations.
