Using the bacterial pentameric ligand‐gated ion channel Gloeobacter violaceus ligand‐gated ion channel, we show that a pair of mutations in Loop Ω (= pre‐β5 strand) inverses the pharmacological effect of crotonate, a ligand of the orthotopic site (homologous to the neurotransmitter binding site).
