RNA profiling of circulating extracellular vesicles (EVs) from blood samples of men undergoing prostate biopsy identifies transcripts associated with clinically significant prostate cancer. Integrative analysis with public tumor datasets links EV‐derived gene signatures to tumor stage and progression‐free survival, highlighting CASP3, XRCC2, and RIT1 ...

Epilepsy is related to theta band brain connectivity and network topology in brain tumor patients
Background Both epilepsy patients and brain tumor patients show altered functional connectivity and less optimal brain network topology when compared to healthy controls, particularly in the theta band.

The Role of the Neurologist after Diagnosis
Psychogenic nonepileptic seizures (PNES) are the most frequent type of paroxysmal events misdiagnosed as epileptic seizures. Neurologists are usually the initial contact in the care of patients with these events, as they are often able to recognize or ...

Bayesian multi-modal model comparison: a case study on the generators of the spike and the wave in generalized spike–wave complexes [PDF]
We present a novel approach to assess the networks involved in the generation of spontaneous pathological brain activity based on multi-modal imaging data.

Effect of Seizures and Epileptiform Discharges on Cognitive Function [PDF]
Summary: Several relationships have been obtained between cognitive impairment and epilepsy‐related or treatment‐related factors. One of these factors is treatment‐related: the central cognitive side effects of the antiepileptic drugs (AEDs). The second and third factors are disease‐related factors, i.e., the effect of the seizures and underlying ...

Intratumour heterogeneity complicates precision management of advanced endometrial cancer. Circulating tumor DNA (ctDNA) offers a minimally invasive strategy to capture tumor evolution and therapeutic resistance. Here, we compare tumor‐agnostic NGS with tumor‐informed ddPCR, outlining their relative sensitivity, concordance, and clinical implications ...

Dynamical intrinsic functional architecture of the brain during absence seizures.
Epilepsy is characterized by recurrent and temporary brain dysfunction due to discharges of interconnected groups of neurons. The brain of epilepsy patients has a dynamic bifurcation that switches between epileptic and normal states.

Interpreting the effects of DNA polymerase variants at the structural level
Using MAVISp and molecular dynamics simulations, we analyzed over 60 000 missense variants in POLE and POLD1 from ClinVar, COSMIC, cBioPortal, and saturation mutagenesis. Identified mechanistic indicators, including stability, binding, and long‐range, enable structural interpretation, providing ACMG‐like evidence for possible reclassification of VUS ...

This study shows that lung adenocarcinomas exploit developmental branching morphogenesis to acquire a therapy resistant basal‐like tumour cell state. This process was found to be regulated by combined TP53 loss‐of‐function and type‐I interferon signalling, identifying a novel axis for biomarker and therapeutic target discovery.
