First tumor microenvironment (TME)‐responsive spermine‐loaded hydrogel with autonomous sensing for advanced local therapy of prostate cancer (PCa) is developed. Spermine can be released from the hydrogel to kill the prostate cancer cells specifically under acidic TME.
