Characterization of an activating factor required for hydrolysis of Gm2 ganglioside catalyzed by hexosaminidase A

open access: closedCanadian Journal of Biochemistry, 1977
Separation of the hexosaminidase A (EC 3.2.1.52) and B isozymes of human liver by ion-exchange chromatography results in recovery of greater than 80% of the activity in crude extracts when synthetic substrates are used to monitor enzyme activity. Only 15% of hexosaminidase activity toward the N-acetylgalactosaminyl (N-acetylneuraminyl) galactosyl ...

Enzyme-linked Immunosorbent Assay for the Ganglioside GM2-Activator Protein. Screening of Normal Human Tissues and Body Fluids, of Tissues of GM2Gangliosidosis, and for Its Subcellular Localization

open access: closedHoppe-Seyler´s Zeitschrift für physiologische Chemie, 1984
An enzyme-linked immunosorbent assay was developed for the quantitation of the human activator protein for ganglioside GM2 degradation by beta-N-acetyl-D-hexosaminidase A. With this assay, various tissues and body fluids of normal subjects and of patients with variant forms of GM2 gangliosidosis were screened for their GM2-activator content.

GM2-Ganglioside Metabolism In Situ in Mucolipidosis IV Fibroblasts

Neurochemical Research, 1999
Mucolipidosis IV (ML IV) is an inherited lysosomal disorder for which the primary biochemical defect has not been identified. In order to detect any defect in glycosphingolipid metabolism, we have examined the metabolism of sphingosine-labeled (3H)G(M2) in situ in fibroblasts from patients diagnosed with ML IV.

Recombinant Ganglioside GM2 Synthase—Expression in Insect Cells and Enzyme Assay

2003
Publisher Summary This chapter investigates the recombinant ganglioside GM2 synthase; and describes the expression of murine β-1,4-N-acetylgalactosaminyltransferase both as a complete membrane-bound enzyme and as a soluble form in the baculovirus insect -cell expression system.

Development of a human monoclonal antibody to ganglioside G(M2) with potential for cancer treatment.

Cancer research, 1997
A human B-lymphoblastoid cell clone, L55-81, that produces human monoclonal antibody (MAb) to ganglioside G(M2) was established from peripheral blood B lymphocytes of a melanoma patient. L55-81 secretes IgMkappa light chain antibody in a serum-free medium.

Vaccination with a bivalent G(M2) and G(D2) ganglioside conjugate vaccine: a trial comparing doses of G(D2)-keyhole limpet hemocyanin.

Clinical cancer research : an official journal of the American Association for Cancer Research, 2001
Immunization with GMK vaccine (G(M2) ganglioside conjugated to keyhole limpet hemocyanin mixed with QS-21 adjuvant) induces anti-G(M2) antibodies in close to 100% of patients. We found previously that anti-G(D2) antibodies could be induced in some patients using G(D2)-keyhole limpet hemocyanin + QS-21 (GDK).
