A peptide library screening platform has been developed to discover covalent transcription factor inhibitors. It identifies an antagonist cysteine residue, which forms an inhibitory disulphide with cJun C269. Conversion of this cysteine to an electrophile generates an irreversible and selective cJun inhibitor, validating this approach for covalent ...

RNA modification is crucial in fibrosis diseases, but the role of m1A modification remains elusive. This study reveals that ALKBH3, an m1A demethylase, drives skin fibrosis by reshaping m6A RNA modification and stabilizing COL1A1 and FN1 mRNAs through METTL3, uncovering a crosstalk between m1A and m6A methylation in pathological events.

Endothelial PD‐1 Regulates Vascular Homeostasis and Oligodendrogenesis during Brain Development
Vascular endothelial cells are fundamental for angiogenesis and brain development. This study reveals that endothelial programmed cell death 1 (PD‐1) is critical for cerebral angiogenesis and blood brain barrier (BBB) maturation. It also mediates interaction with oligodendrocyte precursor cells (OPCs), modulating OPCs differentiation and myelination ...

Macrophage phenotypes are influenced by the microenvironment. Matrix stiffness, a key ECM factor, polarizes macrophage toward a pro‐regenerative phenotype in a 3D physiological environment. CaMKK2 selectively regulates stiffness‐induced macrophage polarization, impacting tumor growth and wound healing.

Prime editors (PEs) enhance gene editing precision but face repair mechanism limitations. DNA methylation (CpG sites) and de novo methyltransferases (DNMT3A/DNMT3B) boost PE efficiency. We developed an episomal PE system (epiPE) using EBNA1/oriP to sustain expression and methylation.
