Abstract GFLV-infected samples from different regions of Iran were subjected to sanitation treatments consisting of meristem-tip culture and thermotherapy. The survey study confirmed the occurrence of GFLV in previously reported areas in Iran. Results also shown the GFLV infection in new regions, e.g. some nurseries located in Karaj and Naghadeh.

The cDNA of the GFLV CP gene amplified from clone pGAB5 by PCR was labeled by photosensitive biotin. The probe sensitivity is 5 pg, and the detectable amount of unlabeled GFLV cDNA was 10 pg by Dot-blot hybridization when the biotin-labeled cDNA was used as a probe.

Grapevine fanleaf virus (GFLV) and Arabis mosaic virus (ArMV), two nepoviruses highly detrimental to grapes, are specifically transmitted by the ectoparasitic nematodes Xiphinema index and X. diversicaudatum, respectively. This specificity of transmission is determined by the coat protein (2CCP).
