SUMMARYSeven hypothyroid women (six postmenopausal and one premenopausal) were treated with 200 or 300 μg of levo‐T4 daily in order to suppress the secretion of TSH and its alpha and beta subunits. After such therapy, serum TSH and beta subunit of TSH (TSH‐β) levels were usually not detectable. Serum alpha decreased from a mean value of 4.5 ± 0.8 ng/ml

Molecular Cloning of the Rhesus Glycoprotein Hormone α-Subunit Gene

A rhesus monkey genomic library was screened with a cDNA for the glycoprotein hormone alpha-subunit. Genomic clones hybridizing with exon-specific probes were selected and the DNA sequences were determined for 1.6 kb of 5'-flanking DNA, all four exons, the second and third introns, all exon-intron junctions, and 357 bp of 3'-flanking DNA.

Efficient preparation of glycoprotein hormones lacking an α-subunit oligosaccharide

The oligosaccharide on alpha-subunit loop 2 (alpha 2) is needed for full glycoprotein hormone efficacy. Efforts to prepare glycoprotein hormone antagonists usually involve removing the alpha 2 oligosaccharide and are hampered by its requirement for efficient heterodimer secretion from mammalian cells.

Serum glycoprotein hormone alpha-subunit levels were determined in 165 patients with pituitary adenomas. Elevated serum alpha-subunit levels were found in 17 patients (acromegaly, 5 of 58; prolactinoma, 6 of 56; nonfunctioning adenoma, 5 of 32; and ACTH-producing adenoma, 1 of 19), most of whom had normal serum TSH and gonadotropin levels.

The amino acid sequence of the alpha subunit of rabbit (lagomorph) lutropin (lLH) has been determined. Overlapping peptides from trypsin and chymotrypsin digestions were isolated by reverse-phase high-pressure liquid chromatography (HPLC). Sequencing was by the dansyl-Edman procedure.
