Unrealized targets in the discovery of antibiotics for Gram-negative bacterial infections
Nature Reviews Drug Discovery, 2023Advances in areas that include genomics, systems biology, protein structure determination and artificial intelligence provide new opportunities for target-based antibacterial drug discovery. The selection of a 'good' new target for direct-acting antibacterial compounds is the first decision, for which multiple criteria must be explored, integrated and ...

Multidrug-resistant Gram-negative bacterial infections
Lancet, TheMultidrug-resistant Gram-negative bacterial infections cause significant morbidity and mortality globally. These pathogens easily acquire antimicrobial resistance (AMR), further highlighting their clinical significance. Third-generation cephalosporin-resistant and carbapenem-resistant Enterobacterales (eg, Escherichia coli and Klebsiella spp ...

Treatment of Gram-negative bacterial infections by potentiation of antibiotics
Current Opinion in Microbiology, 2016Infections caused by antibiotic-resistant pathogens, particularly Gram-negative bacteria, represent significant treatment challenges for physicians resulting in high rates of morbidity and mortality. The outer membrane of Gram-negative bacteria acts as a permeability barrier to many compounds that would otherwise be effective antibacterial agents ...

Gram-negative bacterial infections of the mammary gland in cows
American Journal of Veterinary Research, 1991SUMMARY Naturally acquired gram-negative bacterial intramammary infections (n= 160) were studied in 99 cows over a 2-year period. Escherichia coli, Klebsiella spp, Serratia spp, Enterobacter spp, and unidentified gram-negative bacteria were isolated from 28.8, 39.4, 9.4, 5.0, and 11.2%, respectively, of infected mammary glands.

Combating multidrug-resistant Gram-negative bacterial infections
Expert Opinion on Investigational Drugs, 2013Multidrug-resistant (MDR) bacterial infections, especially those caused by Gram-negative pathogens, have emerged as one of the world's greatest health threats. The development of novel antibiotics to treat MDR Gram-negative bacteria has, however, stagnated over the last half century. This review provides an overview of recent R&D activities in the ...

An update on endotoxin neutralization strategies in Gram-negative bacterial infections
Expert Review of Anti-infective Therapy, 2020Gram-negative bacterial infections represent still a severe problem of human health care, regarding the increase in multi-resistance against classical antibiotics and the lack of newly developed antimicrobials. For the fight against these germs, anti-infective agents must overcome and/or bind to the Gram-negative outer membrane consisting of a ...

Endotoxinʼs role in Gram-negative bacterial infection
Current Opinion in Infectious Diseases, 1995Gram-negative bacterial endotoxin has diverse and profound effects on immunocompetent cells, both in vitro and in vivo. During this past year considerable attention was directed towards defining the mechanisms by which cells recognize endotoxin and in examining the fine structure of the proteins involved. Because of the importance of lipopolysaccharide
