Polygenic Risk for Pre‐Eclampsia and the Long‐Term Risk of Incident Hypertension and Cardiovascular Disease: A Population‐Based Cohort Study

open access: yesBJOG: An International Journal of Obstetrics &Gynaecology, EarlyView.
ABSTRACT Objective To investigate whether genetic predisposition to pre‐eclampsia (PE), measured by a polygenic risk score (PRS), is associated with incident hypertension and cardiovascular disease (CVD) after delivery in Asian women. Design Prospective population‐based cohort study.

Phosphodiesterase‐5 Inhibition and Alzheimer's Disease Risk: A Mendelian Randomisation Study

open access: yesAging Cell, EarlyView.
Preclinical studies suggest PDE5 inhibitors may improve cognition, but observational evidence on Alzheimer's disease (AD) risk is mixed. We performed a cis‐Mendelian randomisation analysis across four AD genome‐wide association studies (GWAS) datasets and a sex‐stratified analysis using UK Biobank data.

Association of Epigenetic Age Acceleration and Mitochondrial DNA‐Based Aging Metrics Provides Insights Into Mechanisms of Aging‐Related Diseases

open access: yesAging Cell, EarlyView.
Older participants exhibit lower mtDNA copy number (CN) and a higher rare heteroplasmy burden, while common heteroplasmy burden remains relatively stable. Higher epigenetic age acceleration is associated with lower mtDNA CN and higher rare heteroplasmy burden in 6316 participants.

The Roles of EDA2R in Ageing and Disease

open access: yesAging Cell, EarlyView.
Elevated expression of the ectodysplasin A2 receptor (EDA2R) has been linked to ageing and disease. We appraise the roles of EDA2R in these processes, exploring mechanisms of action, biomarker potential and therapeutic relevance of EDA2R in multiple contexts. ABSTRACT Ageing is a complex biological process driven, in part, by inflammaging.

PCBP2 Regulates p16INK4a‐Dependent Cellular Senescence in Response to Iron

open access: yesAging Cell, EarlyView.
PCBP2 induces cellular senescence by modulating the expression of p16INK4a and CD40 in response to iron. PCBP2 activates p16INK4a via the functional SNP rs1333046 on the atherosclerosis‐associated CDKN2A/B locus. ABSTRACT Recently, we have identified rs1333046 as one of the candidate functional single nucleotide polymorphisms (fSNPs) on the ...
