Purification of photochemically active halorhodopsin. [PDF]

open access: yesProceedings of the National Academy of Sciences, 1983
We have developed a procedure for the purification of halorhodopsin in a photochemically active state. Solubilization of membranes from a bacteriorhodopsin-negative Halobacterium strain with octyl glucoside was followed by chromatography on hydroxylapatite and octyl-Sepharose gels. All steps were carried out in high-ionic-strength solutions.

Structural Mechanism for Light-driven Transport by a New Type of Chloride Ion Pump, Nonlabens marinus Rhodopsin-3*♦

open access: yesJournal of Biological Chemistry, 2016
The light-driven inward chloride ion-pumping rhodopsin Nonlabens marinus rhodopsin-3 (NM-R3), from a marine flavobacterium, belongs to a phylogenetic lineage distinct from the halorhodopsins known as archaeal inward chloride ion-pumping rhodopsins. NM-R3

Primary charge separation in halorhodopsin [PDF]

open access: yesFEBS Letters, 1985
INTRODUCTION Halorhodopsin (hR) is a retinal-binding protein in the cell membrane of Halobacterium halobium [l-4]. On the basis of light-induced volume change measurements on cell envelope vesicles, hR has been suggested to act as a light-driven Cl- pump [5].

Functional reconstitution of halorhodopsin. Properties of halorhodopsin-containing proteoliposomes.

open access: yesJournal of Biological Chemistry, 1988
A one-step purification method for halorhodopsin was developed. Functional proteoliposomes were prepared from this preparation using cholate, which is removed by dialysis in the presence of asolectin or the polar halobacterial lipids. Light-induced outward directed transport of chloride by halorhodopsin was followed by measuring passive proton efflux ...

FEF inactivation with improved optogenetic methods [PDF]

open access: yes, 2016
Optogenetic methods have been highly effective for suppressing neural activity and modulating behavior in rodents, but effects have been much smaller in primates, which have much larger brains. Here, we present a suite of technologies to use optogenetics

Evaluating the outcomes of pluripotent stem-cell-derived photoreceptor transplantation in retinal repair. [PDF]

open access: yesFEBS J
Transplantation of stem cell‐derived photoreceptors in retinal repair. We summarize methods for differentiating stem cells and describe surgical techniques for transplanting photoreceptor precursors to restore visual function (A). The effectiveness of these techniques is assessed with behavioral (B) and in vitro assays (C) using mouse models that mimic

Optical control of zebrafish behavior with halorhodopsin [PDF]

open access: yesProceedings of the National Academy of Sciences, 2009
Expression of halorhodopsin (NpHR), a light-driven microbial chloride pump, enables optical control of membrane potential and reversible silencing of targeted neurons. We generated transgenic zebrafish expressing enhanced NpHR under control of the Gal4/UAS system.

Noninvasive optical inhibition with a red-shifted microbial rhodopsin [PDF]

open access: yes, 2014
Optogenetic inhibition of the electrical activity of neurons enables the causal assessment of their contributions to brain functions. Red light penetrates deeper into tissue than other visible wavelengths. We present a red-shifted cruxhalorhodopsin, Jaws,
