Pathogenicity of NUSAP1 Variants Is Defined by NMD‐Escape: Evidence From Two Novel Cases and Systematic Population‐Based Variant Analysis

Heterozygous de novo nonsense variants in the penultimate and last exons of NUSAP1 were identified in two unrelated individuals, predicted to escape NMD. In population data, nonsense variants were observed in exons 1–9 (of 11) in NUSAP1 but were absent from its 3′‐terminal region.

Early On‐Treatment Change in SALT as a Pragmatic Predictor of Ritlecitinib Response in Moderate‐To‐Severe Alopecia Areata: A Real‐World Cohort Study

Ritlecitinib induced rapid clinical improvement in moderate‐to‐severe alopecia areata in real‐world practice. Early change in SALT at Week 12 strongly discriminated Week‐24 responders, supporting a pragmatic treat‐to‐target checkpoint, alongside improvements in clinician‐reported and patient‐reported outcomes and a favorable safety profile.

Management of chronic urticaria: Current status and future prospect

Chronic urticaria is driven by mast cell activation through autoimmune, inflammatory, and neuroimmune pathways. A structured approach combining diagnosis, patient‐reported outcomes, stepwise therapy, treatment optimization, monitoring and emerging targeted agents may improve disease control and enable more personalized management.
