Yeast Complementation Assays Demonstrating the Importance of the Affinity Tag Position in Membrane Protein Purification, as Exemplified by HpUreI, the pH‐Gated Urea Channel of Helicobacter pylori

Affinity tags help to purify proteins, but can interfere with protein function or structure. Yeast complementation assays to study Helicobacter pylori's UreI protein are used. Results indicate that the tag position and residual amino acids resulting from tag cleavage have a significant influence on protein permeability to urea and ammonia.
