A modified Spalteholz technique with preservation of the histology
Annals of Anatomy - Anatomischer Anzeiger, 2001The Spalteholz technique for producing clear macroscopic specimens was introduced 100 years ago, and is still in use today because it immediately provides transparent three-dimensional specimens. The classical clearing procedure with peroxide severely damages formalin-fixed tissue and inhibits further investigation on a histological level.

HISTOLOGICAL TECHNIQUES FOE THE ELECTRON MICROSCOPE
Journal of the Royal Microscopical Society, 1951SYNOPSIS.A technique for cutting thin sections for the electron microscope is described. Adequate fixation of the tissue is one of the limiting factors. The tissue is fixed in osmium tetroxide, embedded in Ester wax, and sectioned on a Cambridge rocking microtome. The technique has been used to examine various normal rat tissues.
