Reconstructing enzyme evolution by protein engineering

open access: yesFEBS Letters, EarlyView.
Natural enzyme evolution can be retraced by protein engineering methods such as directed evolution, rational design, and ancestral sequence reconstruction. These approaches reveal how enzymes emerged from ligand‐binding scaffolds, developed varying substrate preferences, formed oligomeric complexes, adapted to environmental changes, and evolved novel ...

Le numérique et le dictionnaire : l’impact de l’artéfact numérique sur la lexicographie (1970-2010)

open access: yesHumanités Numériques
Dictionaries, taken in the broad sense of lexicographical works, have undergone a number of changes due to the emergence of computer technology. Initially (1970-1990), these developments took two directions: (1) the transposition of classic dictionaries ...

Identification of a Shiga toxin A‐derived peptide internalized into Gb3 receptor‐bearing cells via interaction with the Shiga toxin B subunit

open access: yesFEBS Letters, EarlyView.
The process of internalization of the Shiga toxin A subunit via formation of a complex with the Shiga toxin B subunit, which specifically binds to the Gb3 receptor. The peptide is designed to act as a carrier of drugs into cancer cells. Here, we explored the potential of peptides derived from the catalytic A subunit of Shiga toxin (STxA) to be drug ...

Reconstructing the invention of the wheel using computational structural analysis and design

open access: yesRoyal Society Open Science
The invention of the wheel is widely credited as a pivotal moment in human history, yet the details surrounding its discovery are shrouded in mystery. There remains no scholarly consensus on key questions such as where, how and by whom this technology ...

Investigating transcription factor dynamics in health and disease using FRAP

open access: yesFEBS Letters, EarlyView.
FRAP analysis of GFP‐tagged transcription factors reveals how molecular mobility and target engagement change in response to drug treatment. By combining live‐cell imaging, quantitative model fitting, and statistical analysis, this approach uncovers transcription factor dynamics linked to disease mechanisms, providing a powerful framework for ...

Conserved binding mode but diverse interfaces of MreC‐PBP2 interactions

open access: yesFEBS Letters, EarlyView.
The crystal structure of abMreC reveals a conserved two β‐barrel architecture and provides structural insights into its role within the bacterial elongasome. The abMreC–abPBP2 complex model identifies the molecular basis of MreC‐mediated PBP2 recognition, contributing to the regulation of peptidoglycan synthesis.

Identification of the plant mitochondrial OrfX protein: A mass spectrometry approach

open access: yesFEBS Letters, EarlyView.
The mitochondrial genome of plants contains an open reading frame, orfx, which encodes a rare protein that has so far escaped mass spectrometric detection. The protein resembles the c‐subunit of bacterial twin‐arginine‐motif‐dependent protein translocases (TatC).

An epithelial GPR35 isoform supports tumor‐associated transcriptional and metabolic phenotypes

open access: yesFEBS Letters, EarlyView.
GPR35 generates two functionally distinct isoforms with previously unresolved roles. GPR35‐short mediates immune‐cell chemotaxis, while GPR35‐long is enriched in colorectal cancer epithelium, where it supports increased metabolism, proliferation, and tumor‐associated transcriptional programs.

L’impératif de la sobriété numérique : enjeux et définitions

open access: yesHumanités Numériques
This article outlines the history of digital technology, identifying the main trends at work and documenting their ecological implications. It then draws up an overview of the main regulations at work, and concludes that they have more to do with ...

The role of miR‐335‐5p in the redifferentiation of BRAF p.V600E thyroid cancers

open access: yesMolecular Oncology, EarlyView.
The BRAF p.V600E mutation promotes thyroid cancer dedifferentiation and radioiodine resistance. Using a network approach, we identified miR‐335‐5p as a key regulator of BRAF‐mutated thyroid tumors. Restoring miR‐335‐5p increased thyroid‐specific gene expression and iodine uptake in cells and organoids.
