Eukaryotic DNA Polymerases in Homologous Recombination.

open access: yesAnnual Review of Genetics, 2016
Homologous recombination (HR) is a central process to ensure genomic stability in somatic cells and during meiosis. HR-associated DNA synthesis determines in large part the fidelity of the process.

Biosecurity Primitive: Polymerase X‐based Genetic Physical Unclonable Functions

open access: yesAdvanced Science, EarlyView.
What if every cell line can carry an unforgeable genomic fingerprint—one that proves its origin, ensures its authenticity, and thwarts unauthorized duplication? Here, CRISPR‐induced DNA lesions paired with TdT‐mediated repair produce robust, unique, and unclonable genetic signatures—advancing the concept of molecular Physical Unclonable Functions (PUF)

The many facets of homologous recombination at telomeres

open access: yesMicrobial Cell, 2015
The ends of linear chromosomes are capped by nucleoprotein structures called telomeres. A dysfunctional telomere may resemble a DNA double-strand break (DSB), which is a severe form of DNA damage. The presence of one DSB is sufficient to

Cell cycle-dependent control of homologous recombination.

open access: yesActa Biochimica et Biophysica Sinica, 2017
DNA double-strand breaks (DSBs) are among the most deleterious type of DNA lesions threatening genome integrity. Homologous recombination (HR) and non-homologous end joining (NHEJ) are two major pathways to repair DSBs.

Preface-Homologous Recombination

open access: yes, 2021
DNA double-strand breaks (DSBs) are the most harmful lesions to DNA in the cell. To cope with these insults, all organisms have devised two main types of evolutionary conserved mechanisms for their repair, homologous recombination (HR), and non-homologous end joining (NHEJ).

A High‐Fidelity RNA‐Targeting Cas13X Downregulates Connexin43 in Macroglia: A Novel Neuroprotective Strategy for Glaucoma

open access: yesAdvanced Science, EarlyView.
This study highlights the role of connexin43 (Cx43) in mediating glia activation and neuroinflammation in glaucoma. Using Cx43 knockout mice, it is shown that macroglial cells‐derived Cx43 modulates microglial activation, proliferation, migration, and polarization.

Spatiotemporal dynamics of homologous recombination repair at single collapsed replication forks

open access: yesNature Communications, 2018
How factors involved in homologous recombination interact and function is a matter of interest. Here the authors use super-resolution imaging to describe the spatiotemporal dynamics of proteins associated with homologous recombination DNA repair in ...

Comprehensive Characterization of Bihormonal Cells and Endocrine Cell Lineages in Mammalian Pancreatic Islets

open access: yesAdvanced Science, EarlyView.
This study integrates dual‐reporter genetics, imaging flow cytometry, and single‐cell sequencing to characterize rare bihormonal cells in mouse and human pancreatic islets. Gcg⁺Ins⁺ cells resemble α‐cells rather than transitional states. Cross‐species gene network analysis refines islet cell taxonomy and reveals human‐specific δ‐cell subtypes ...

Enhancing Specificity, Precision, Accessibility, Flexibility, and Safety to Overcome Traditional CRISPR/Cas Editing Challenges and Shape Future Innovations

open access: yesAdvanced Science, EarlyView.
CRISPR/Cas9, while transformative, faces challenges in specificity, precision, delivery, accessibility, flexibility, and safety. This review addresses these limitations by highlighting strategies to reduce off‐target effects, exploring HDR‐based and alternative editing approaches, and evaluating advanced delivery mechanisms.

Induction of homologous recombination between sequence repeats by the activation induced cytidine deaminase (AID) protein

open access: yeseLife, 2014
The activation induced cytidine deaminase (AID) protein is known to initiate somatic hypermutation, gene conversion or switch recombination by cytidine deamination within the immunoglobulin loci.
