Nitrilase Catalyzes Amide Hydrolysis as Well as Nitrile Hydrolysis

While amides were reported to be completely inert as substrates for all nitrilases reported to date, the nitrilase from Rhodococcus rhodochrous J1, which catalyzes the hydrolytic cleavage of the C-N triple bond in nitrile to form acid and ammonium, was surprisingly found to catalyze hydrolysis of amide to acid and ammonium stoichiometrically.

Hydrolysis and protection from hydrolysis of enkephalins in human plasma

Seven groups of enkephalin-degrading enzymes and three groups of inhibitors active on these enzymes were separated from human plasma. The activity of the enzymes in hydrolyzing enkephalins and of the inhibitors in protecting enkephalins from proteolysis was measured.

The glycosidic bond might be degraded by chemical and/or enzymatic agents. Comparative studies revealed that chemical hydrolysis is nonspecific and on the other hand the enzymatic is regiospecific and stereospecific. The glycosides are chemically susceptible to acid conditions and only in some cases to basic conditions.

Abstract There are two general approaches to the use of hydrolysis and esterification in asymmetric synthesis, and usually both involve the use of enzymes as asymmetric catalysts. In one case the asymmetric catalyst is used in a kinetic resolution process in which one enantiomer of a racemic mixture reacts more rapidly than its antipode.
