SenExo‐cCCT2 Reprograms Senescence Response and Anti‐Tumor Immunity Following FOLFIRINOX Chemotherapy in Pancreatic Ductal Adenocarcinoma

open access: yesAdvanced Science, EarlyView.
This study demonstrates that SenExo‐cCCT2 enables targeted delivery of cCCT2 to pancreatic cancer cells, where it disrupts IPO13‐mediated nuclear translocation of UBC9, impairing SUMOylation‐dependent DNA damage repair and inducing senescence. Senescent tumor cells secrete CXCL10 to recruit CD8⁺ T‐cells while upregulating PD‐L1.

FOXM1 Protects Against Myocardial Ischemia‐Reperfusion Injury in Rodent and Porcine Models by Suppressing MKRN1‐Dependent LKB1 Ubiquitination

open access: yesAdvanced Science, EarlyView.
FOXM1 maintains mitochondrial bioenergetic function by inhibiting MKRN1‐mediated ubiquitination of LKB1 in cardiomyocytes. Loss of FOXM1 in cardiomyocytes results in upregulation of MKRN1, which enhances LKB1 ubiquitination and disrupts AMPK signaling and energy metabolism pathways. Conversely, FOXM1 overexpression preserves mitochondrial bioenergetics

A Charge‐Adhesive Targeted DNA Gel Bandage for the Precision Treatment of Inflammatory Bowel Disease

open access: yesAdvanced Science, EarlyView.
DNA gel bandage (DNAgb) is designed to precisely target the site of inflammation of IBD, by achieving dual specific localization of inflamed mucosa through electrostatic interactions and ApITGA4‐mediated affinity targeting. DNAgb effectively the activation of inflammatory pathways and reshapes the immune microenvironment.

CRISPR Screening Reveals a Novel Role for FOXH1 in Regulating Pluripotency of Porcine Embryonic Stem Cells

open access: yesAdvanced Science, EarlyView.
This study performs dual CRISPR screens in pEPSCs. Fitness screening identifies essential genes for viability, while FACS‐based screening uncovers core pluripotency factors. It highlights FOXH1 as a key regulator that maintains the stem cell state through a dual‐action mechanism: preserving pluripotency by enhancing chromatin accessibility and ...

Rationally Designed TadA‐Derived Cytosine Editors Enable Context‐Independent Zebrafish Genome Editing

open access: yesAdvanced Science, EarlyView.
A next‐generation cytosine base editor (zTadCBE) and other variants are developed by combining the high‐efficiency (TadCBEa) and low‐indel (TadCBEMax) architectures, followed by the introduction of zebrafish‐optimized mutations (V82S, Q154R) in the TadA domain.
