A Rationally Engineered Spleen‐Tropic One‐Component Lipid‐mRNA Complex (OncoLRC) for Cancer Vaccines

OncoLRC, a one‐component lipid‐mRNA complex, enables efficient spleen‐targeted delivery at an exceptionally low lipid‐to‐mRNA mass ratio (1.5:1), robustly activates immune responses, inhibits tumor growth, and synergizes with checkpoint blockade, presenting a next‐generation platform for mRNA vaccines.

Corticosteroids improve the efficacy of immune checkpoint blockade therapy in metastatic murine breast cancer. By normalizing the tumor immune microenvironment, corticosteroids reduce immunosuppressive signals, restore T‐cell function, and promote antitumor immune responses, resulting in enhanced tumor control.

Advanced Human Immune Cell‐Organoid Co‐Cultures for Functional Testing of Cancer Nanovaccines

Pancreatic ductal adenocarcinoma remains difficult to treat. We establish an organoid–immune co‐culture using patient‐derived organoids and matched T‐cells to assess cancer vaccines. A mesothelin‐targeted nanovaccine activates antigen‐specific T‐cells, increases IFN‐γ, and targets MSLN+ organoids.
