Emestrin‐Type Epidithiodiketopiperazines Inhibited Gasdermin D‐Mediated Pyroptosis via Caspase‐3/7 Activation

open access: yesMedComm, Volume 7, Issue 1, January 2026.
This study demonstrates that emestrin‐type ETPs suppress GSDMD‐mediated pyroptosis through caspase‐3/7 activation, thereby attenuating inflammatory responses and conferring protection against sepsis. In THP‐1 macrophages, ETPs significantly reduced pyroptosis and secretion of the proinflammatory cytokines IL‐1β and IL‐18.

Prenatally Diagnosed 7q11.23 Copy Number Variations: A Retrospective Case Series

open access: yesMolecular Genetics &Genomic Medicine, Volume 14, Issue 1, January 2026.
Prenatal 7q11.23 CNVs exhibit 100% ultrasound anomalies in deletions (cardiovascular defects, growth restriction) and 50% in duplications. Inherited CNVs (50% deletions, 43% duplications) correlate with milder outcomes, while de novo variants drive high TOP rates (76.5%), emphasizing genomic testing and parental studies for precise counseling. Knowledge Gaps and Controversies on Cardiopulmonary Exercise Testing in the Assessment of Pulmonary Vascular Disease: An Official Statement of the Pulmonary Vascular Research Institute Exercise and Right Ventricular Function Task Force

open access: yesPulmonary Circulation, Volume 16, Issue 1, January 2026.
ABSTRACT Exercise testing has long been essential for evaluating diagnosis, prognosis, and functional status in pulmonary hypertension (PH). Recent advances have clarified its role in defining reference values and prognostic markers. Nonetheless, substantial knowledge gaps persist regarding the implementation of invasive cardiopulmonary exercise ...

Manipulation of Wnt/β‐Catenin Signaling by Synthetic Frizzled Agonist and LRP Antagonist in Organoid Cultures and In Vivo

open access: yesSmall Methods, Volume 10, Issue 2, 22 January 2026.
Harnessing synthetic key receptor modulators, this study presents a breakthrough in Wnt/β‐catenin regulation. The FZD agonist RRP‐pbFn bypasses LRP5/6 to drive organoid expansion and tissue regeneration, while the LRP antagonist RRP‐Dkk1c eradicates tumors. These tools enable precise pathway control, advancing regenerative and cancer therapies.
