Inosine monophosphate dehydrogenase activity in acute leukaemia
Leukemia Research, 1987Inosine monophosphate dehydrogenase (IMPD) is an important enzyme in de-novo purine synthesis. The level of IMPD activity has been suggested to determine whether acute leukaemia cells proliferate (if the activity is high) or differentiate (if IMPD activity is low).

Rationally Designed Inhibitors of Inosine Monophosphate Dehydrogenase
Journal of Medicinal Chemistry, 1997Functionalized 2-alkyl derivatives of inosinic acid have been synthesized to serve as reversible as well as irreversible inhibitors of the human type II enzyme inosine monophosphate dehydrogenase. These compounds were designed to react with Cys-331 of the enzyme to form covalent bonds so as to interfere with the normal enzyme mechanism which involves ...

Inhibition of inosine monophosphate dehydrogenase by sesquiterpene lactones
Biochimica et Biophysica Acta (BBA) - General Subjects, 1987Inosine monophosphate (IMP) dehydrogenase had previously been determined to be a likely target enzyme for the sesquiterpene lactones, a class of potential anti-neoplastic drugs. IMP dehydrogenase was purified approx. 770-fold from the P-388 lymphocytic leukemia tumor cell line.

Inosine Monophosphate Dehydrogenase Polymorphisms and Renal Allograft Outcome
Transplantation, 2012Interindividual variation in inosine monophosphate dehydrogenase (IMPDH) enzyme activity and adverse effects caused by mycophenolate mofetil (MMF) inhibition may be genetically determined, and if so, transplant recipients should receive personalized dosing regimens of MMF, which would maximize efficacy and minimize toxicity.

Novel guanidine-Based inhibitors of inosine monophosphate dehydrogenase
Bioorganic & Medicinal Chemistry Letters, 2002A series of novel guanidine-based small molecule inhibitors of inosine monophosphate dehydrogenase (IMPDH) was explored. IMPDH catalyzes the rate determining step in guanine nucleotide biosynthesis and is a target for anticancer, immunosuppressive and antiviral therapy.

Studies on inosine monophosphate dehydrogenase. Steady state kinetics
Biochimica et Biophysica Acta (BBA) - Enzymology, 1976The reaction catalyzed by IMP dehydrogenase (IMP: NAD+ oxidoreductase EC 1.2.1.14) from Aerobacter aerogenes has been investigated kinetically at pH 8.1 as a three reactant system by means of steady-state velocity studies in the absence of products, as well as by inhibition studies using products and substrate analogues.

A novel process of inosine 5′-monophosphate production using overexpressed guanosine/inosine kinase
Applied Microbiology and Biotechnology, 1997A novel process for producing inosine 5'-monophosphate (5'-IMP) has been demonstrated. The process consists of two sequential bioreactions; the first is a fermentation of inosine by a mutant of Corynebacterium ammoniagenes, and the second is a unique phosphorylating reaction of inosine by guanosine/inosine kinase (GIKase).

Inosine 5’-Monophosphate Dehydrogenase as a Chemotherapeutic Target
1995Inosine 5’-monophosphate, EC 1.1.1.205 (IMPDH) catalyzes the conversion of IMP to XMP utilizing NAD as a proton acceptor. Its role in catalyzing the rate determining step in the biosynthesis of GTP1 gives IMPDH a position of central importance in cellular activity because of the myriad activities of GTP in biosynthesis and cellular regulation.

Inosine monophosphate dehydrogenase inhibition: mycophenolate mofetil
2001Over the past two decades, the development of new immunosuppressive drugs and the prevention of post-transplant infection have substantially improved the survival of transplanted patients and of organ allografts. However, the prevention of acute rejection and consequently of chronic allograft dysfunction, as well as that of long-term metabolic ...

A new enzyme-mediated synthesis of 2-vinylinosine, a compound with broad-spectrum RNA antiviral activity, is described. In order to understand the mechanism of action of this compound, we synthesized its monophosphate and investigated the behavior of that compound toward the enzyme, inosine monophosphate dehydrogenase (IMPDH), a key enzyme involved in ...
