Influence of external K+ on potassium efflux in isolated chicken enterocytes

1. Efflux of K+ was measured in pre-loaded (86Rb+) chicken enterocytes incubated in buffers with external K+ concentration ([K+]0) between 1 and 40 mM. 2. A decrease in [K+]0 from 6 to 1 mM reduced the rate constant of K+ efflux, whereas it was stimulated by increasing [K+]0 from 6 to 40 mM. 3.

Effects of anisosmotic buffers on K+ transport in isolated chicken enterocytes

Cells isolated by hyaluronidase incubation from chicken small intestine were used to study the effects of anisosmotic buffers on K+ transport. Hypo-osmolarity (200 mosmol.l-1) reduced both the ouabain-sensitive and the ouabain-resistant, but bumetanide-sensitive, net K+ influx and increased the K+ efflux.

Cl−/HCO 3 − exchanger is operative in isolated enterocytes from rat jejunum

Enterocytes isolated from rat jejunum were tested for the existence of a Cl-/HCO(-3) exchange, previously evidenced in basolateral membrane vesicles but not in brush border. Cells were found to retain functional integrity and transport capabilities long enough to allow Cl- fluxes to be measured.

[Method of isolation of intact enterocytes from rat duodenal mucosa].

A procedure is suggested for obtaining intact enterocytes from rat duodenum mucosa by the tissue trypsinization in the isotonic Krebs solution (pH 7.9). The maximum yield of certain enterocytes is reached at the enzyme concentration of 0.03%; the incubation time--15 min; 200-250 mg of mucosa produces 3,7 X 10(6)--4,7 X 10(6) cells.
