β-淀粉样肽对海马 CA1 区神经元的 短暂性外向 K+电流的影响
目的: 为了在离子通道水平探讨β-淀粉样肽( β-AP)在 Alzheimer 病中的作用。方法: 用 7~ 21 d 大鼠,以链霉蛋 白酶 E 酶解加吸管吹打法制备海马 CA1区神经元; 利用全细胞膜片钳技术, 观察了β-AP25~ 35对急性分离海马 CA1 区锥体细 胞的短暂性 K+电流的影响。结果: β-AP25~ 35对短暂性 K+电流有明显抑制作用, 且呈明显浓度依赖性、时间依赖性和部分电 压依赖性。结论: β-AP 对 K+通道的影响在 Alzheimer 病的发生机制中具有重要作用 ...
