We evaluated the efficiency of a minimal sampling approach for DNA metabarcoding and developed a novel single‐amplicon workflow. We compared the diet and assessed geographic and seasonal variations in the isotopic niches of red‐footed boobies across three Indian Ocean atolls. Our results demonstrate plasticity in both diet and trophic niche, likely driven by local environmental conditions, breeding stages, and colony‐specific factors.
