Optimizing photoexcitation conditions for time‐resolved X‐ray solution scattering experiments

open access: yesFEBS Open Bio, EarlyView.
Time‐resolved X‐ray solution scattering (TR‐XSS) is a powerful technique to visualize how proteins change their structure in real time after light activation. Selecting the right laser photoexcitation conditions—fluence, excitation geometry, and sample refresh rate—is critical to maximize the experimental signal while avoiding unwanted side effects ...

Purification and preparation of Marchantia polymorpha Auxin Response Factor 2 for phase separation studies

open access: yesFEBS Open Bio, EarlyView.
We describe detailed protocols for the purification and preparation of Marchantia polymorpha Auxin Response Factor 2 (MpARF2). This protein is fused to an MBP solubility tag and an mNG fluorescent tag and is purified from Escherichia coli. The presented procedures make it possible to study MpARF2 assemblies, which could arise from phase separation ...

Protocol for quantifying miRNA trafficking across the endosomal membrane

open access: yesFEBS Open Bio, EarlyView.
An in vitro protocol measures miRNA uptake into endosomes isolated from mammalian cell extracts, which are free of subcellular contaminants. Performed at 37 °C in the presence of ATP, it ensures the import of single‐stranded miRNA into the endosomal lumen.

Identification and characterisation of calcitonin receptor isoforms expressed in glioblastoma derived glioma stem and U‐87 MG cells

open access: yesFEBS Open Bio, EarlyView.
Glioblastoma cells express calcitonin receptor variants (CT receptor isoforms) that may help them survive stress. Using qPCR, transcript‐specific long‐read nanopore sequencing, immunofluorescence co‐localisation and comparative sequence analysis, this study identifies a novel alternatively spliced CALCR transcript that encodes the CTb receptor isoform ...

Exploration of the pyramid complex of King Djedkare: season 2018 [PDF]

open access: yesPražské Egyptologické Studie, 2019
During the latest fieldwork in the pyramid complex of King Djedkare in the 2018 season, the Egyptian mission focused oncleaning and documentation of the central and northern parts of the king’s funerary temple, including the open courtyard (T.e), and the

Comparative assessment of crystallographic and cryo‐EM models in the Protein Data Bank

open access: yesFEBS Open Bio, EarlyView.
Raw data obtained by X‐ray crystallography or cryo‐EM result in experimental maps, ultimately fitted by atomic models. Although the physical principles are different, the final results can be viewed, compared, and evaluated in the same way. With cryogenic electron microscopy (cryo‐EM) on track to surpass X‐ray crystallography as the preferred method ...

Association of Pelvic Lymph Nodes Inclusion with Late Side Effects in Prostate Cancer Patients Undergoing Curative Radiotherapy

open access: yesBulletin of Urooncology
Objective: Curative radiotherapy is one of the two leading definitive treatment options for prostate cancer, along with surgery. The inclusion of pelvic lymph nodes in curative radiotherapy for prostate cancer is controversial.

Submicroscopic changes of secretory cardiomyocytes in the intermediate and late postcompression period of crush syndrome

open access: yesMorphologia
Background. With the onset of compression, in response to stress, pain syndrome, and heart overload, myoendocrinocytes of the heart secrete atrial natriuretic peptide, which regulates blood pressure, glucose and lipid metabolism, slows down the processes of lipid peroxidation, prevents hypertrophy of cardiomyocytes, the development of fibrosis ...

Structural and biochemical insights into the thermostable esterase Ta0887 from Thermoplasma acidophilum

open access: yesFEBS Open Bio, EarlyView.
In this study, a novel esterase from the thermoacidophilic archaeon Thermoplasma acidophilum was biochemically and structurally characterized. Our results demonstrate that Ta0887 is a highly thermostable esterase that preferentially hydrolyzes p‐nitrophenyl hexanoate and possesses an α‐helical cap domain that likely contributes to its substrate ...
