Live-attenuated influenza virus vaccine strain with an engineered temperature-sensitive and genetically stable viral polymerase variant

open access: yesJournal of Virology
Vaccination against seasonal influenza is considered an effective means of reducing morbidity and mortality. Live-attenuated vaccines offer more protection against influenza than inactivated vaccines as they efficiently induce cellular immunity and ...

Live attenuated chimeric porcine circovirus vaccine

open access: yes, 2014
The present invention provides a novel chimeric porcine circovirus infectious DNA clone and live attenuated chimeric virus with the PCV2, preferably of subtype PCV2b, capsid gene integrated into a non-pathogenic PCV1 virus genome.

NS1‐Induced PARP1 Phase Transitions Prolongs Host Transcriptional Pausing to Promote Influenza A Virus Replication

open access: yesAdvanced Science, EarlyView.
After IAV infection, NS1 induces PARP1 phase transition, reducing molecular mobility in condensates, prolonging host transcriptional pause, aiding RdRp cap snatching, and promoting viral transcription. ABSTRACT Influenza A virus (IAV) commandeers host transcriptional machinery to replicate, but mechanisms remain elusive.

Current status of the development of dengue vaccines

open access: yesVaccine: X
Dengue fever is caused by the mosquito-borne dengue virus (DENV), which is endemic in more than 100 countries. Annually, there are approximately 390 million dengue cases, with a small subset manifesting into severe illnesses, such as dengue haemorrhagic ...

Engineered Allosteric Biosensor for In Situ DNA Glycosylase Detection in Neuroblastoma Risk Stratification and Drug Resistance Monitoring

open access: yesAdvanced Science, EarlyView.
An engineered allosteric biosensor (UUU‐DZ‐tFNA) was developed, in which UDG specifically recognizes and activates a DNAzyme to achieve signal cycle amplification in a sequentially activatable mode. UUU‐DZ‐tFNA achieves in situ UDG detection across multiple biological levels and facilitates risk stratification and drug resistance monitoring of NB ...

Optimal immunization strategies for Saanen goats against goatpox

open access: yesBMC Veterinary Research
Background Goatpox is a highly contagious disease caused by the Capripoxvirus, primarily affecting sheep and goats. Breeds like the Saanen are particularly vulnerable, especially in enzootic areas, and face risks not only from the disease itself but also

Sarbecoviruses Suppress Innate Immunity by Recruiting PPM1A to Modulate STAT2 and ORF9b Phosphorylation

open access: yesAdvanced Science, EarlyView.
Sarbecovirus ORF9b recruits host phosphatase PPM1A to suppress innate immunity through dual mechanisms. PPM1A directly dephosphorylates ORF9b to sustain its activity and indirectly downregulates STAT2 phosphorylation to impair interferon signaling. Pharmacological inhibition of PPM1A restores antiviral responses and limits sarbecovirus replication ...

Optimizing Cancer Vaccinations Using a Physiologically‐Based Pharmacokinetic/Pharmacodynamic (PBPK/PD) Model

open access: yesAdvanced Science, EarlyView.
Where and when should a cancer vaccine be administered? A whole‐body PBPK model demonstrates that systemic immune transport critically determines vaccination efficacy. By simulating how organ‐specific immune interactions influence treatment response, the study provides a quantitative framework for optimizing cancer vaccination strategies.

An Adenoviral‐Vectored Tp0326 Vaccine Elicits Robust Functional Antibodies to Prevent Treponema pallidum Dissemination in a Rabbit Model

open access: yesAdvanced Science, EarlyView.
Vaccination promotes cellular infiltration into primary lesions and effectively inhibits T. pallidum dissemination to distal organs. Notably, transfer of lesion tissue from immunized animals fails to establish infection in naive recipients, confirming specific protective immunity.
