Spatial Transcriptomics Reveals Transcriptomic and Immune Microenvironment Reprogramming during Thyroid Carcinoma Dedifferentiation

Spatial transcriptomic analysis of Anaplastic thyroid carcinoma (ATC)‐differentiated thyroid carcinoma (DTC) coexisting tumors reveals that coexisting DTC regions with ATC‐like genomic alterations undergo sequential transcriptomic reprogramming and immune microenvironment remodeling, ultimately evolving into a full ATC pathological phenotype.

TREM2‐Mediated Cholesterol Efflux in Macrophages Inhibits Anti‐Tumor Immunity via Limitation of CD4+ T and NK Cells

Impaired anti‐tumor immunity is mediated by TREM2 + macrophages in non‐small cell lung cancer. TREM2 + macrophages prevent anti‐tumor immunity depending on the limitation of CD4 + T and NK cells. TREM2 promotes cholesterol efflux via ABCA1 to limit the production of CX3CL1 in macrophages.

METTL10‐Mediated PIAS3 Methylation Links Purine Metabolism to Gastric Cancer Progression

METTL10‐mediated methylation of PIAS3 at K442 inhibits its SUMO E3 ligase activity toward MITF. This reduces MITF SUMOylation and subsequent RNF4‐mediated ubiquitination, leading to MITF protein accumulation and gastric cancer progression. Senescence‐Inducing Therapy Sequential NIR‐II Mild Photothermal/Senolytic Therapy of Triple Negative Breast Cancer

Targeting triple‐negative breast cancer remains challenging due to limited therapies and drug resistance. A sequential theranostic strategy combining palbociclib‐induced senescence (enhancing heat sensitivity via HSP70 downregulation) with an NIR‐II nanoplatform (IQ NPs) for mild photothermal therapy (PTT) and quercetin‐mediated senolysis is developed.
