Extracellular Vesicle‐Packaged circTAX1BP1 from Cancer‐Associated Fibroblasts Regulates RNA m6A Modification through Lactylation of VIRMA in Colorectal Cancer Cells

This study reveals that CAFs‐derived EVs enriched with circTAX1BP1 drive CRLM by promoting VIRMA lactylation and SP1/TGF‐β signaling, creating a positive feedback loop via ITGA11+ myCAFs. Targeting EV‐circTAX1BP1 and TGF‐β disrupts this loop, offering a novel therapeutic strategy for CRLM.

HmmyCAFs may form a triple immunosuppressive niche: possibly secrete ECM (POSTN, etc.) as physical barriers to block CD8⁺ T cells, induce CD8⁺ T exhaustion via those molecules, and use HIF‐1α‐driven metabolism to create acidic, nutrient‐poor microenvironments that suppress T cells.
