Interpreting the effects of DNA polymerase variants at the structural level
Using MAVISp and molecular dynamics simulations, we analyzed over 60 000 missense variants in POLE and POLD1 from ClinVar, COSMIC, cBioPortal, and saturation mutagenesis. Identified mechanistic indicators, including stability, binding, and long‐range, enable structural interpretation, providing ACMG‐like evidence for possible reclassification of VUS ...

Ixazomib inhibits proteasome‐mediated degradation of topoisomerase I induced by irinotecan, thereby restoring drug sensitivity and promoting tumor cell death in colorectal cancer. Irinotecan, a topoisomerase I (topoI) inhibitor, is widely used for colorectal cancer, but resistance remains a major clinical challenge.

Gender and collective action: A conceptual framework for analysis [PDF]
"This paper presents a framework for investigating the intersection of collective action and gender; i.e. how gender-oriented analysis can foster more effective collective action in the context of agriculture and natural resource management and how ...

Dormant cancer cells can hide in distant organs for years, evading treatment and the immune system. This review highlights how signals from the surrounding tissue and immune environment keep these cells inactive or trigger their reawakening. Understanding these mechanisms may help develop therapies to eliminate or control dormant cells and prevent ...

RURAL DEVELOPMENT IN SERBIA AND THE LOCAL COMMUNITIES
Rural areas encompass the most part of Serbia and they are very heterogenous in relation to natural conditions for development, production capacities, employment, infrastructure, income and standard of living.

LOCAL ACTION GROUPS, A SOLUTION TO THE RURAL DEVELOPMENT OF ROMANIA [PDF]
For Romania, the rural area represent a difficult problem because it has a high share. During the communist era, a forcefully reduction of the rural share was undertaken through various means, most often destructive (Ailenei, 2001).

This study shows that lung adenocarcinomas exploit developmental branching morphogenesis to acquire a therapy resistant basal‐like tumour cell state. This process was found to be regulated by combined TP53 loss‐of‐function and type‐I interferon signalling, identifying a novel axis for biomarker and therapeutic target discovery.

Amenable actions of locally compact groups
AbstractWe consider a locally compact group G with jointly continuous action G × Z → Z on a locally compact space. The finite Radon (regular Borel) measures M(G) act naturally on various function spaces supported on Z, including the continuous bounded functions CB(Z).

Combining osimertinib with the STING agonist ADU‐S100 activates innate and adaptive immunity to overcome the non‐inflamed microenvironment of Egfr‐mutant lung cancer. This combination increases NK and CD8+ T‐cell infiltration, associated with activation of the STING‐IRF3 pathway and local immunogenic cell death.

Loss of IGF‐1R impairs DNA‐PKcs recruitment to chromatin leading to defective end‐joining
IGF‐1R promotes radioresistance by facilitating DNA‐PKcs recruitment to chromatin, enabling non‐homologous end‐joining (NHEJ) repair of double‐strand breaks. Inhibition or loss of IGF‐1R disrupts this recruitment to damage sites, driving compensatory reliance on microhomology‐mediated end‐joining (MMEJ) repair.
