Deciphering and Targeting the Schwannoma‐Neuron‐Macrophage Crosstalk for the Treatment of Schwannomatosis and Associated Pain

open access: yesAdvanced Science, EarlyView.
We established patient‐derived SWN cell lines and orthotopic PDX models that recapitulate patient pain phenotypes, alongside a novel intravital DRG imaging platform to track macrophage infiltration and neuronal pain responses. Using these models, we define HMGB1–CCL2–IL‐6 signaling crosstalk driving pain and identify EGF signaling as a key regulator of

Functional asymmetry of the spine in standing and sitting positions [Funkční asymetrie páteře vstoje a vsedě]

open access: yesActa Universitatis Palackianae Olomucensis. Gymnica, 2010
BACKGROUND: A sedentary lifestyle, together with functional asymmetry of the body, are potentially dysfunctional factors. OBJECTIVE: In this project, the authors are trying to identify the connections between a sedentary lifestyle and the amount of ...

Relationships Between Muscle Activation and Thoraco-Lumbar Kinematics in Direction-Specific Low Back Pain Subgroups During Everyday Tasks

open access: yesBiomechanics
Background/Objectives: The assessment of relationships between trunk muscle activity and thoraco-lumbar movements during sagittal bending has demonstrated that low back pain (LBP) subgroups (flexion pattern and active extension pattern motor control ...

CRISPLD2 Attenuates Intervertebral Disc Degeneration by Suppressing Oxidative Stress‐Induced Ferroptosis through the miR‐548I‐IL17A Axis

open access: yesAdvanced Science, EarlyView.
This study identifies CRISPLD2 as a key protector against IVDD. By regulating ferroptosis through the CRISPLD2–miR‐548I–IL17A axis, CRISPLD2 maintains NPCs homeostasis and reduces oxidative stress. Restoring CRISPLD2 expression effectively alleviates disc degeneration and highlights a promising therapeutic strategy for discogenic low back pain ...

Comparison of postoperative complications and outcomes following primary versus revision discectomy: A national database analysis

open access: yesJournal of Craniovertebral Junction and Spine
Background: Lumbar microdiscectomy is a surgical procedure that is frequently used in the treatment of symptomatic lumbar herniation. Differences in outcomes following primary and revision lumbar microdiscectomy have been previously studied, with reports

Textiloma mimicking soft tissue tumor: an uncommon complication of posterior lumbar surgery

open access: yesPAMJ Clinical Medicine, 2020
Textiloma (from Greek textile and ome = tumour) also called gossypiboma (from Latin Gossypium = cotton and Swahili boma = hiding place) is defined as the inadvertent retention of textile foreign body after surgical intervention leading to various ...

Sirt3 Genetically Engineered Apoptotic Bodies Alleviate Skeletal Aging by Limiting Aggravated NLRP3 Inflammasome Activation of Senescent Macrophages

open access: yesAdvanced Science, EarlyView.
The senescence of macrophages leads to a decrease in the expression of Sirtuin3 (Sirt3), which in turn aggravates the NLRP3 inflammasome activation and IL‐1β secretion, thereby promoting the aging‐related osteoporosis. Sirt3‐enriched apoptotic bodies (ABs‐Sirt3) generated through genetic editing approaches can alleviate skeletal aging by targeting ...

Sensory Nerve‐Derived CGRP Controls Osteoclastogenesis by Limiting Macrophage Bioenergetics in Bone Repair

open access: yesAdvanced Science, EarlyView.
Sensory nerves help bones heal. This study shows that the neuropeptide CGRP, released from sensory nerves, slows down macrophage energy production, preventing excessive bone breakdown. By revealing this nerve–immune–metabolism connection, the work provides new insight into how the body balances bone repair and opens doors to targeted treatments ...

Elevated TRIM25 Impairs Poly (ADP‐ribose) Metabolism via PARG Degradation and Mediates Compression‐Induced Intervertebral Disc Degeneration

open access: yesAdvanced Science, EarlyView.
TRIM25 acts as a multifunctional hub driving intervertebral disc degeneration under mechanical stress. Mechanical compression significantly upregulates TRIM25 expression, establishing it as a key E3 ubiquitin ligase platform. TRIM25 targets PARG and Ku80 via distinct molecular interfaces, triggering their ubiquitination and degradation.
