Endogenous Ceramide 24:1 Constrains Th17‐Driven Neutrophilic Inflammation by Antagonizing EP2 Signaling

Cer24:1 levels are reduced in neutrophilic asthma and inversely correlate with disease severity and airway neutrophilia. Restoring Cer24:1 suppresses pathogenic Th17 differentiation by engaging EP2 on CD4+ T cells, thereby dampening the JAK2–STAT3–RORγt axis and reducing IL‐17 production.

TGM2 Aggravates Acute Pancreatitis by Impairing Macrophage Efferocytosis Through Inhibition of the STAT6–GAS6 Axis

This study shows TGM2 is upregulated in AP, impairing macrophage efferocytosis by inhibiting the STAT6–GAS6 axis. The lactoferrin‐modified, ROS‐responsive LF‐LNP@si‐TGM2 targets pancreatic macrophages, silences TGM2, restores the axis, and alleviates AP.

Hepatocyte BDNF Acts as a Novel Immune Checkpoint to Restrain TLR4‐Mediated Acute Hepatitis

This study identifies hepatocyte‐derived BDNF as an endogenous TLR4 antagonist that alleviates acute hepatitis. BDNF is downregulated in hepatocytes via REST‐mediated transcriptional repression during ALI/ALF. Mechanistically, BDNF binds to TLR4 on macrophages to suppress inflammation.

Kinesin‐Induced Buckling Reveals the Limits of Microtubule Self‐Repair

This study shows that kinesin‐driven buckling induces extensive microtubule lattice damage that often exceeds intrinsic self‐repair and leads to filament failure. While curvature, motor motility, and force individually cause limited damage, their combination overwhelms repair.

In Vivo CRISPR Screening Identifies the Glutamate Receptor GRIA2 as Promoting Peritoneal Metastasis of Gastric Cancer via Calcium‐Dependent β‐Catenin Activation

We identified GRIA2 as a critical driver of gastric cancer peritoneal metastasis through in vivo CRISPR screening. Mechanistically, GRIA2‐mediated calcium influx inhibits GSK3β and activates Wnt/β‐catenin signaling, driven by glutamate from cancer‐associated fibroblasts.
