Genomics‐led approach to drug testing in models of undifferentiated pleomorphic sarcoma

open access: yesMolecular Oncology, EarlyView.
GA text Genomic data from undifferentiated pleomorphic sarcoma patients and preclinical models were used to inform a targeted drug screen. Selected compounds were tested in 2D and 3D cultures of UPS cell lines. A combination of trametinib and infigratinib was synergistic in the majority of UPS cell lines tested, which was further confirmed in an ex ...

Maximum Approximated Likelihood Estimation [PDF]

open access: yesarXiv, 2019
Empirical economic research frequently applies maximum likelihood estimation in cases where the likelihood function is analytically intractable. Most of the theoretical literature focuses on maximum simulated likelihood (MSL) estimators, while empirical and simulation analyzes often find that alternative approximation methods such as quasi-Monte Carlo ...

Unraveling LINE‐1 retrotransposition in head and neck squamous cell carcinoma

open access: yesMolecular Oncology, EarlyView.
The novel RetroTest method allows the detection of L1 activation in clinical samples with low DNA input, providing global L1 activity and the identification of the L1 source element. We applied RetroTest to a real‐world cohort of HNSCC patients where we reported an early L1 activation, with more than 60% of T1 patients showing L1 activity.

BMP antagonist CHRDL2 enhances the cancer stem‐cell phenotype and increases chemotherapy resistance in colorectal cancer

open access: yesMolecular Oncology, EarlyView.
Overexpression of CHRDL2 in colon cancer cells makes them more stem‐like and resistant to chemo‐ and radiotherapy. CHRDL2‐high cells have upregulation of the WNT pathway, genes involved in the DNA damage response (DDR) pathway and epithelial‐to‐mesenchymal transition (EMT). This leads to quicker repair of damaged DNA and more cell migration.

The Maximum Likelihood Degree of Farlie Gumbel Morgenstern Bivariate Exponential Distribution [PDF]

open access: yesarXiv
The maximum likelihood degree of a statistical model refers to the number of solutions, where the derivative of the log-likelihood function is zero, over the complex field. This paper examines the maximum likelihood degree of the parameter in Farlie-Gumbel-Morgenstern bivariate exponential distribution.

Simultaneous inhibition of TRIM24 and TRIM28 sensitises prostate cancer cells to antiandrogen therapy, decreasing VEGF signalling and angiogenesis

open access: yesMolecular Oncology, EarlyView.
TRIM24 and TRIM28 are androgen receptor (AR) coregulators which exhibit increased expression with cancer progression. Both TRIM24 and TRIM28 combine to influence the response of castrate‐resistant prostate cancer (CRPC) cells to AR inhibitors by mediating AR signalling, regulation of MYC and upregulating VEGF to promote angiogenesis. Castrate‐resistant

Parsimonious and Efficient Likelihood Composition by Gibbs Sampling [PDF]

open access: yesarXiv, 2015
The traditional maximum likelihood estimator (MLE) is often of limited use in complex high-dimensional data due to the intractability of the underlying likelihood function. Maximum composite likelihood estimation (McLE) avoids full likelihood specification by combining a number of partial likelihood objects depending on small data subsets, thus ...
