The [2Fe‐2S] cluster of mitochondrial outer membrane protein mitoNEET has an O2‐regulated nitric oxide access tunnel

open access: yesFEBS Letters, EarlyView.
The mitochondrial outer membrane iron–sulphur ([Fe‐S]) protein mitoNEET is a target of the type‐2 diabetes drug pioglitazone. Its unknown molecular function is linked to respiratory complex activity and mitochondrial function. We discovered that O2 protects the mitoNEET [2Fe‐2S] cluster against NO oxidation and desensitization towards reduction by H2S.

Fast crystallization of rotating membrane proteins [PDF]

open access: yesPhys. Rev. Lett. 123, 148101 (2019), 2019
We examine the interactions between actively rotating proteins moving in a membrane. Experimental evidence suggests that such rotor proteins, like the ATP synthases of the inner mitochondrial membrane, can arrange themselves into lattices. We show that crystallization is possible through a combination of hydrodynamic and repulsive interactions between ...

Proteomics of Cytochrome c Oxidase-Negative versus -Positive Muscle Fiber Sections in Mitochondrial Myopathy [PDF]

open access: yes, 2019
The mosaic distribution of cytochrome c oxidase(+) (COX+) and COX - muscle fibers in mitochondrial disorders allows the sampling of fibers with compensated and decompensated mitochondrial function from the same individual.

A histidine‐rich extension of the mitochondrial F0 subunit ATP6 from the ice worm Mesenchytraeus solifugus increases ATP synthase activity in bacteria

open access: yesFEBS Letters, EarlyView.
The glacier ice worm Mesenchytraeus solifugus survives year‐round at 0 °C. Its ATP6 subunit, which forms a regulatory component of the proton pore in mitochondrial ATP synthase, has a carboxy‐terminal extension not found in any other organism examined to date. Here, we show that fusion of this extension to the homologous AtpB protein in E. coli results

RNAi-based gene silencing in primary mouse and human adipose tissues

open access: yesJournal of Lipid Research, 2007
Cultured adipocyte cell lines are a model system widely used to study adipose function, but they exhibit significant physiological differences compared with primary cells from adipose tissue.

BNIP3 and NIX mediate Mieap-induced accumulation of lysosomal proteins within mitochondria. [PDF]

open access: yesPLoS ONE, 2012
Mieap, a p53-inducible protein, controls mitochondrial quality by repairing unhealthy mitochondria. During repair, Mieap induces the accumulation of intramitochondrial lysosomal proteins (designated MALM for Mieap-induced accumulation of lysosome-like ...

The apo LETM1 F‐EF‐hand adopts a closed conformation that underlies a multi‐modal sensory role in mitochondria

open access: yesFEBS Letters, EarlyView.
We present the first solution structure of the Ca2+‐depleted LETM1 F‐EF‐hand through a D676A/N678A Ca2+ binding‐deficient mutant, revealing a closed hydrophobic cleft caused by a unique F1‐helix pivot. The apo LETM1 F‐EF‐hand exhibits regiospecific hot and cold unfolding, sensitivity to physiological pH changes and potential for promiscuous heterotypic

Taurine promotes glucagon‐like peptide‐1 secretion in enteroendocrine L cells

open access: yesFEBS Letters, EarlyView.
Taurine, a sulfur‐containing amino acid, is likely taken up by enteroendocrine L cells via the taurine transporter. This process increases the levels of cytosolic ATP. The increase in intracellular Ca2+ concentrations and glucagon‐like peptide‐1 secretion through membrane depolarization is caused by the closure of ATP‐sensitive potassium channels ...

Generation of a mitochondrial protein compendium in Dictyostelium discoideum

open access: yesiScience, 2022
Summary: The social ameba Dictyostelium discoideum has emerged as a powerful model to study mitochondrial genetics and bioenergetics. However, a comprehensive inventory of mitochondrial proteins that is critical to understanding mitochondrial processes ...

A Biophysical Model of the Mitochondrial ATP-Mg/Pi Carrier [PDF]

open access: yes, 2012
Mitochondrial adenine nucleotide (AdN) content is regulated through the Ca2+-activated, electroneutral ATP-Mg/Pi carrier (APC). The APC is a protein in the mitochondrial carrier super family that localizes to the inner mitochondrial membrane (IMM).
