We use an engineered mutant of the protein nanopore Mycobacteria smegmatis porin A (MspA) to examine single stranded DNA. The ssDNA is either held stationary within the pore by a biotin-streptavidin anchor or is processively drawn through the pore by a phi29 DNA polymerase.
