Single‐molecule DNA flow‐stretch assays for high‐throughput DNA–protein interaction studies

open access: yesFEBS Open Bio, EarlyView.
We describe an optimised single‐molecule DNA flow‐stretch assay that visualises DNA–protein interactions in real time. Linear DNA fragments are tethered to a surface and stretched by buffer flow for fluorescence imaging. Using λ and φX174 DNA, this protocol enhances reproducibility and accessibility, providing a versatile approach for studying diverse ...

A human-system integration framework and its application for special vehicle interface design under typical human readiness levels

open access: yesiScience
Summary: Life cycle Human System Integration (HSI) practices are crucial for optimizing human system performance, reducing costs, and ensuring safety. To address the limited HSI practices under typical Human Readiness Levels (HRLs), our study proposes an

The natural selection of conservative science [PDF]

open access: yesStudies in History and Philosophy of Science Part A, 2019
Social epistemologists have argued that high risk, high reward science has an important role to play in scientific communities. Recently, though, it has also been argued that various scientific fields seem to be trending towards conservatism -- the increasing production of what Kuhn (1970) would have called `normal science'.

Exon 7 splicing of ERα predicts poor prognosis and increases phenotypic heterogeneity in luminal a subtype breast cancer

open access: yesFEBS Open Bio, EarlyView.
ERα splice variant ERα∆7 lacks the C‐terminus, and its expression may change phenotypes of breast cancers. Our results showed that ERα∆7 is found in the luminal A subtype, and elevated ERα∆7 levels are linked to improved cell survival with lower proliferation and migration.

Polarization-insensitive narrowband reflective wavefront manipulation through all-dielectric anisotropic subwavelength structures

open access: yesiScience
Summary: Local metasurfaces provide high flexibility in shaping wavefronts but suffer from poor frequency selectivity. Here, we numerically present a reflective all-dielectric metasurface platform achieving simultaneous local phase control and spectral ...

Screening and epitope characterization of Nidogen‐2‐specific nanobodies

open access: yesFEBS Open Bio, EarlyView.
Camel immunization and phage display were employed to generate high‐affinity VHH nanobodies against Nidogen‐2. After library construction, biopanning, ELISA screening, sequencing, and recombinant expression, selected nanobodies were purified and characterized, leading to the preliminary exploration of a nanobody‐based sandwich ELISA for specific ...

Quantitative proteomic analysis reveals different characteristics of bladder cancer cells after exposure to bisphenol A

open access: yesFEBS Open Bio, EarlyView.
Bisphenol A (BPA), a common chemical in plastics, exerts dual effects on bladder cancer cells: low doses promote growth and migration, while high doses suppress growth and migration. Multi‐omics and bioinformatics reveal BPA acts via MAPK and inflammatory pathways.

Laser-induced voltage of table salt for deep ultraviolet pulsed laser detection

open access: yesiScience
Summary: To meet the requirements of fast response and simple process of deep ultraviolet (UV) pulsed laser detector, table salt (TS) was used as laser detection material in combination with a variable resistor to achieve single-pulse laser detection ...

Rapid screening of staphylokinase protein variants using an unpurified cell‐free expression system

open access: yesFEBS Open Bio, EarlyView.
An unpurified cell‐free protein synthesis (CFPS) platform enables rapid functional screening of staphylokinase variants. Direct plasminogen‐activation assays performed in microplate format provide real‐time activity readouts, allowing rapid identification and ranking of variants with improved or reduced fibrinolytic activity without protein ...
