Vaccines based on Nef and on Nef/ΔV2 Env

Microbes and Infection, 2005
Modified vaccinia virus Ankara (MVA) is a potent vaccine vector, which proved its safety, immunogenity and efficacy in preclinical and clinical studies. The rational for the development of a vaccine against HIV based on the regulatory protein Nef delivered by MVA combined with a V2-deleted Env protein is discussed.

The Nef Reaction of Isocyanides

Synthesis, 2014
The interaction of isocyanides with acyl chlorides was first reported by Nef in 1892. The resulting α-keto imidoyl chlorides may be hydrolyzed to form α-keto amides. Relatively few data are available on this reaction and the chemistry of α-keto imidoyl chlorides has been largely underestimated until some recent uses in intra- and intermolecular ...

Nef—an adaptor adaptor?

Trends in Cell Biology, 1998
The Nef protein of human immunodeficiency virus (HIV) is a crucial factor in viral pathogenesis. One of the roles of this multifunctional protein is to decrease the cell-surface expression of CD4, a component of the virus receptor complex, and of MHC class I molecules.

Nef: out and in?

Nature Immunology, 2006
Lentiviral Nef protein has many functions in the evasion of host immune defenses. Another 'chink' in the host immune 'armor' has now been exposed by Nef's ability to inhibit immunoglobulin class switching.

Nef and Transcription

Science, 2004
STKE The Nef protein encoded by human immunodeficiency virus (HIV) appears to function in pathogenesis through interactions in a signaling complex associated with the T cell receptor. Witte et al. identified an unexpected interaction partner of Nef: the protein Eed (embryonic ectodermal development factor).

The CD4 Determinant for Downregulation by HIV-1 Nef Directly Binds to Nef. Mapping of the Nef Binding Surface by NMR

Biochemistry, 1996
Using heteronuclear NMR spectroscopy, we demonstrate that a 13-residue peptide (MS-QIKRLLSEKKT) from the cytoplasmic tail of CD4 binds to Nef protein. This part of CD4 is critical for downregulation of CD4 by HIV-1 Nef [Aiken et al. (1994) Cell 76, 853-864]. We show that a control peptide without the central dileucine does not bind to Nef.
