A Knowledge‐Guided Graph Learning Approach Bridging Phenotype‐ and Target‐Based Drug Discovery

open access: yesAdvanced Science, Volume 12, Issue 16, April 24, 2025.
Knowledge‐Guided Drug Relational Predictor (KGDRP), a graph representation learning approach, effectively integrates multiple omics data, including biological network data, gene expression data, and sequence data that incorporates chemical molecular structures.

Zonated Copper‐Driven Breast Cancer Progression Countered by a Copper‐Depleting Nanoagent for Immune and Metabolic Reprogramming

open access: yesAdvanced Science, EarlyView.
Spatially resolved multiomics reveals that high‐copper zones are closely associated with tumor metastasis, immune‐desert phenotypes, and oxidative‐phosphorylation (OXPHOS)‐dependent proliferation. A targeted bacterial nanovesicle delivering copper‐depleting agents effectively reduces tumor copper levels, inhibits OXPHOS‐dependent tumor growth, and ...

Osteoblast‐Derived Mitochondria Formulated with Cationic Liposome Guide Mesenchymal Stem Cells into Osteogenic Differentiation

open access: yesAdvanced Science, Volume 12, Issue 12, March 27, 2025.
This study demonstrates that the transfer of mitochondria from osteoblasts to mesenchymal stem cells (MSCs) enhances osteogenic differentiation. MSCs receiving osteoblast‐derived mitochondria exhibit increased expression of key osteogenic markers (Runt‐related transcription factor 2, RUNX2; Osterix, OSX; Osteopontin, OPN) and activate the BMP2 (Bone ...

DNA‐PKcs‐Driven YAP1 Phosphorylation and Nuclear Translocation: a Key Regulator of Ferroptosis in Hyperglycemia‐Induced Cardiac Dysfunction in Type 1 Diabetes

open access: yesAdvanced Science, EarlyView.
In the context of chronic hyperglycemia, a DDR is initiated, leading to the pathological activation of DNA‐PKcs in the diabetic heart. This activated DNA‐PKcs directly interacts with and phosphorylates YAP1 at Thr226, thereby increasing the nuclear expression of YAP1.
