Liquid biopsy epigenetics: establishing a molecular profile based on cell‐free DNA
Cell‐free DNA (cfDNA) fragments in plasma from cancer patients carry epigenetic signatures reflecting their cells of origin. These epigenetic features include DNA methylation, nucleosome modifications, and variations in fragmentation. This review describes the biological properties of each feature and explores optimal strategies for harnessing cfDNA ...

Intein‐based modular chimeric antigen receptor platform for specific CD19/CD20 co‐targeting
CARtein is a modular CAR platform that uses split inteins to splice antigen‐recognition modules onto a universal signaling backbone, enabling precise, scarless assembly without re‐engineering signaling domains. Deployed here against CD19 and CD20 in B‐cell malignancies, the design supports flexible multi‐antigen targeting to boost T‐cell activation and

Development of therapies targeting cancer‐associated fibroblasts (CAFs) necessitates preclinical model systems that faithfully represent CAF–tumor biology. We established an in vitro coculture system of patient‐derived pancreatic CAFs and tumor cell lines and demonstrated its recapitulation of primary CAF–tumor biology with single‐cell transcriptomics ...

Drill Tool Design Based on Fatigue Failure of BHA in S-shaped Well
Wellbore curvature can cause fatigue damage to BHA in the S-shaped complex trajectory borehole, which could seriously impact the life of drilling tools. In view of this, BHA in the S-shaped wellbore has been mechanically analyzed.

The ADS general-purpose optimization program [PDF]
The mathematical statement of the general nonlinear optimization problem is given as follows: find the vector of design variables, X, that will minimize f(X) subject to G sub J (x) + or - 0 j=1,m H sub K hk(X) = 0 k=1,l X Lower I approx less than X sub I

We developed and validated a DNA methylation–based biomarker panel to distinguish pleural mesothelioma from other pleural conditions. Using the IMPRESS technology, we translated this panel into a clinically applicable assay. The resulting two classifier models demonstrated excellent performance, achieving high AUC values and strong diagnostic accuracy.
