An efficient early‐pooling protocol for environmental DNA metabarcoding
Environmental DNA, Volume 4, Issue 6, Page 1212-1228, November 2022., 2022 Our results show that an early‐pooling protocol with post‐1st‐PCR exonuclease purification and an increased amount of DNA template will reduce the risk of tag jumping, the costs for consumables and reagents, and the handling time in the library preparation and that it produces comparable results to a 2nd‐PCR‐indexing protocol.

Validation using independent test datasets showed that the true positive rate of our CRC diagnostic model increased substantially as CRC progressed from stage I to stage II.
