Oxaloacetate permeation in rat kidney mitochondria: Pyruvate/oxaloacetate and malate/oxaloacetate translocators

The mechanism of oxaloacetate efflux from rat kidney mitochondria has been investigated in view of its possible role both in gluconeogenesis and in transferring cytosolic reducing equivalents into mitochondria. Thus reconstruction of the malate/oxaloacetate shuttle made possible by the oxaloacetate carrier has been made.

Reconstruction, using non-synaptosomal rat brain mitochondria, has been made here of the malate/oxaloacetate shuttle, made possible by the occurrence in rat brain mitochondria of a carrier mediated transport for oxaloacetate (Km = 60 microM), sensitive to dicarboxylate analogues and mersalyl, and of the two isoenzymes of malate dehydrogenase.

The formation of alanine in the transamination of α-amino-n-butyrate and glutamate with oxaloacetate in presence of liver homogenates has been studied. It could be quantitatively explained in terms of pyruvate formed by the decarboxylation of oxaloacetate and subsequent transamination.

Abstract The complete vibrational spectra of dimethyl oxaloacetate and dimethyl oxaloacetate-d2 have been recorded and analyzed. The i.r. spectra were recorded at liquid N2 as well as ambient temperature. Tentative vibrational assignments are proposed based on an enol structure in the crystalline phase.
